摘要
目的观察黄芩苷联合白细胞介素-7(IL-7)对小鼠肝纤维化模型中肝脏组织病理形态的影响。方法用四氯化碳(CCL_(4))诱导形成小鼠肝纤维化模型。将50只昆明小鼠随机等量分成5组,分别是模型组、黄芩苷组、黄芩苷联合IL-7组、IL-7组、对照组。各干预组(黄芩苷组、黄芩苷联合IL-7组、IL-7组)于造模第3周首日开始予黄芩苷进行灌胃25 mg每日2次和(或)肌肉注射IL-710μg隔日1次;第6周末,取出小鼠肝组织,采用HE染色、网状纤维染色及Masson染色观察各组肝组织炎症及纤维化情况,TUNEL检测各组肝细胞凋亡情况,并采用Ishak评分系统对小鼠肝组织炎症活动度及纤维化程度进行评分。结果成功建立小鼠肝纤维化模型;肝组织病理显示,与模型组比较,各干预组的肝组织炎症活动度及肝纤维化呈不同程度减轻,其中黄芩苷联合IL-7组比黄芩苷组或IL-7组的肝组织炎症及纤维化改善更明显。黄芩苷联合IL-7组与模型组的肝组织炎症及纤维化程度比较差异均有统计学意义(均P<0.05)。与模型组比较,各干预组中肝组织的肝细胞凋亡也有减少,黄芩苷联合IL-7干预小鼠肝纤维化模型后可明显减少组织中肝细胞的凋亡(P<0.05)。结论黄芩苷联合IL-7可改善CCL_(4)诱导的小鼠肝纤维化过程中肝脏组织炎症,阻断纤维化形成,且两者间可能存在协同作用。
Objective To observe the pathological effect of baicalin combined with interleukin-7(IL-7)on liver tissue in mice with hepatic fibrosis induced by carbon tetrachloride(CCl_(4)).Methods A liver fibrosis model was induced in mice using CCL_(4).Fifty Kunming mice were randomly divided into five groups:model group,baicalin group,baicalin+IL-7 group,IL-7 group,and control group.From the third week of modeling,the intervention groups received either baicalin(25 mg twice daily by gavage),IL-7(10μg intramuscularly every other day),or both.At the end of week 6,liver tissue was collected,and histological changes were assessed by hematoxylin and eosin(HE)staining,reticular fiber staining,and Masson staining.Hepatocyte apoptosis was evaluated by TUNEL assay,and inflammation activity and fibrosis scores were assessed using the Ishak scoring system.Results A successful mouse model of liver fibrosis was established.Histopathological analysis showed that,compared to the model group,all intervention groups exhibited reduced hepatic inflammation and fibrosis,with the baicalin+IL-7 group demonstrating more significant improvement than the baicalin or IL-7 monotherapy groups.Significant differences in hepatic inflammation and fibrosis were observed between the baicalin+IL-7 group and the model group(P<0.05).Additionally,hepatocyte apoptosis was reduced in the intervention groups compared to the model group,with a significant decrease observed in the baicalin+IL-7 group(P<0.05).Conclusion Baicalin combined with IL-7 can improve hepatic inflammation and inhibit fibrosis progression in the CCl_(4)-induced liver fibrosis model in mice,with potential synergistic effects.
