摘要
目的探讨厚朴“发汗”过程中厚朴酚与和厚朴酚的含量变化趋势。方法采用高效液相色谱(HPLC)法测定彭州20年树龄及雅安20年、40年树龄厚朴“发汗”过程中厚朴酚与和厚朴酚的含量。色谱柱为Wondasil C18柱(150 mm×4.6 mm,5μm),流动相为甲醇-水(80∶20,V/V),流速为1.0 mL/min,检测波长为294 nm,柱温为30℃,进样量为10μL。结果厚朴酚与和厚朴酚的质量浓度分别在0.0061~0.1940 mg/mL、0.0018~0.0560 mg/mL范围内与各自峰面积线性关系良好(r≥0.9998,n=6);检测限分别为0.00097,0.00224 mg/mL,定量限分别为0.01210,0.00350 mg/mL;精密度、稳定性、重复性试验结果的RSD均小于2.0%;平均回收率分别为98.79%,99.29%,RSD分别为1.71%,1.20%(n=6)。彭州和雅安20年树龄厚朴中厚朴酚与和厚朴酚含量在“发汗”第7天达峰值,分别为3.48%,3.83%;雅安40年树龄厚朴中厚朴酚与和厚朴酚含量在第5天达峰值,为12.14%。结论所建立的方法快速高效、准确度高、专属性强,可用于厚朴中厚朴酚与和厚朴酚的含量测定。树龄是厚朴“发汗”过程中厚朴酚与和厚朴酚含量变化的主要影响因素。
Objective To investigate the changing trend of of magnolol and honokiol content during the"Fahan"process of Magnolia officinalis.Methods High-performance liquid chromatography(HPLC)method was used for the content determination of magnolol and honokiol during the"Fahan"process of 20-year-old Magnolia officinalis from Pengzhou and 20-year-old and 40-year-old Magnolia officinalis from Ya′an.The chromatographic column was Wondasil C18 column(150 mm×4.6 mm,5μm),the mobile phase was methanol water(80∶20,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 294 nm,the column temperature was 30℃,and the injection volume was 10μL.Results The linear ranges of magnolol and honokiol were 0.0061-0.1940 mg/mL and 0.0018-0.0560 mg/mL(r≥0.9998,n=6),respectively.The limits of detection were 0.00097 mg/mL and 0.00224 mg/mL,respectively,and the limits of quantification were 0.01210 mg/mL and 0.00350 mg/mL,respectively.The RSDs of precision,stability,and repeatability test results were all lower than 2.0%.The average recoveries of magnolol and honokiol were 98.79%and 99.29%,with RSDs of 1.71%and 1.20%(n=6),respectively.The content of magnolol and honokiol in 20-year-old Magnolia officinalis from Pengzhou and Ya′an reached its peak value on the 7th day of"Fahan"(3.48%and 3.83%,respectively).The content of magnolol and magnolol in 40-year-old Magnolia officinalis from Ya′an reached its peak value on the 5th day(12.14%).Conclusion The established method is quick,efficient,accurate,and highly specific,which can be used for the content determination of magnolol and honokiol in Magnolia officinalis.The age of trees is the main influencing factor of the changes in magnolol and honokiol in Magnolia officinalis during the″Fahan″process.
