摘要
目的探讨幽门螺杆菌(Hp)感染胃癌癌组织混合性谱系激酶1(MLK1)、果蝇zeste基因增强子同源物2(EZH2)、性别决定相关基因簇2(SOX2)、轴蛋白(Axin)表达水平与病理学特征的关系。方法选取2020年6月-2023年6月山东第一医科大学附属人民医院收治的102例胃癌合并Hp感染患者为感染组,同期收治的26例胃癌未合并Hp感染患者为未感染组。采集癌组织及癌旁组织,检测标本中MLK1、EZH2、SOX2、Axin蛋白表达,比较两组癌组织及癌旁组织中蛋白表达阳性率,对比不同病理参数胃癌合并Hp感染患者癌组织中蛋白表达阳性率。结果与癌旁组织比,感染组和未感染组癌组织中MLK1、EZH2蛋白表达阳性率更高(P<0.05),SOX2、Axin更低(P<0.05);与未感染组比,感染组癌组织和癌旁组织中MLK1、EZH2蛋白表达阳性率更高(P<0.05),SOX2、Axin更低(P<0.05)。Ⅲ~Ⅳ期、肌层/浆膜层、低分化、有淋巴结转移的胃癌合并Hp感染患者癌组织中MLK1、EZH2蛋白表达阳性率高于Ⅰ~Ⅱ期、黏膜层/黏膜下层、中高分化、无淋巴结转移者(P<0.05),SOX2、Axin低于Ⅰ~Ⅱ期、黏膜层/黏膜下层、中高分化、无淋巴结转移者(P<0.05)。结论Hp感染胃癌患者癌组织MLK1、EZH2蛋白高表达,SOX2、Axin低表达,与TNM分期、浸润深度、分化程度及淋巴结转移有关。
OBJECTIVE To explore the expressions of mixed lineage kinase 1(MLK1),enhancer of zeste homolog 2(EZH2),sex-determining-region Y-box 2(SOX2)and axin in cancer tissues of the gastric cancer patients with Helicobacter pylori infection and analyze the association with clinicopathologic features.METHODS Totally 102 gastric cancer patients complicated with H.pylori infection who were treated in Shandong First Medical University Affiliated People′s Hospital from Jun.2020 to Jun.2023 were assigned as the infection group,meanwhile,26 gastric cancer patients without H.pylori infection were chosen as the no infection group.The cancer tissues and para-carcinoma tissues were collected to detect the expressions of MLK1,EZH2,SOX2 and Axin proteins.The positive rates of expressions of the proteins in the cancer tissues and para-carcinoma tissues were compared between the two groups.The positive rates of expressions of the proteins in the cancer tissues were compared among the gastric cancer patients complicated with H.pylori infection who had different pathological parameters.RESULTS The positive rates of expressions of MLK1 and EZH2 proteins of the infection group and the no infection group were higher in the cancer tissues than in the para-carcinoma tissues(P<0.05),the positive rates of expressions of SOX2 and Axin were lower in the cancer tissues than in the para-carcinoma tissues(P<0.05).The positive rates of expressions of MLK1 and EZH2 in cancer tissues and para-carcinoma tissues were higher in the infection group than in the no infection group(P<0.05),the positive rates of expressions of SOX2 and Axin were lower in the infection group than in the no infection group(P<0.05).The positive rates of expressions of MLK1 and EZH2 in cancer tissues of the gastric cancer patients complicated with H.pylori infection who hadⅢtoⅣstage,myenteric/submucosal layer,low differentiation and lymph node metastasis were higher than those of the patients who hadⅠtoⅡstage,myenteric/submucosal layer,intermediate to high differentiation and did not have lymph node metastasis(P<0.05).The positive rates of expressions of SOX2 and Axin of the patients who hadⅢtoⅣstage,myenteric/submucosal layer,low differentiation and lymph node metastasis were lower than those of the patients who hadⅠtoⅡstage,myenteric/submucosal layer,intermediate to high differentiation and did not have lymph node metastasis(P<0.05).CONCLUSION The gastric cancer patients with H.pylori infection show high expressions of MLK1 and EZH2 proteins in cancer tissues and low expressions of SOX2 and Axin,which are associated with the TNM stage,depth of infiltration,degree of differentiation and lymphatic metastasis.
