摘要
【目的】本研究旨在探究卷丹与不同百合种及品种间的杂交亲和性,以期培育出优良性状的卷丹杂交后代。【方法】将卷丹与7个百合种及品种进行远缘杂交,首先采用液体培养基悬滴法测定父本百合的花粉生活力,杂交后统计杂交组合的果实膨大率,并对获得的杂交蒴果进行胚拯救以获得杂种苗。【结果】实验结果表明,7种百合花粉在9号萌发液中(150 g/L蔗糖,200 mg/L硼酸,75 mg/L氯化钙)普遍萌发较好,其中龙牙百合、‘Strawberry Event’的花粉萌发率高于其他父本品种,花粉活力较好。卷丹与亚洲百合杂种系品种远缘杂交的果实膨大率较高,其中与‘Strawberry Event’杂交的果实膨大率最高,为14.7%,并且只有此杂交组合成功获得了2株杂交后代。【结论】本研究表明卷丹与亚洲百合杂种系杂交亲和性更好,为今后卷丹的远缘杂交育种提供了技术支持。
【Objective】The purpose of this study is to explore the hybridization compatibility between the Lilium lancifolium and different lily species and varieties in order to produce the hybrid offspring with excellent traits.【Method】Lilium lancifolium Thunb.was remotely bred with 7 lily species and varieties.First,the pollen vitality of the paternal lily was measured by the pollen liquid medium method,and the fruit expansion rate of the hybrid combination was counted after crossing,and the hybrid capsule was obtained by embryo rescue to obtain hybrid seedlings.【Result】The results showed that 7 kinds of lily powder germinated well in No.9 germination solution(150 g/L sucrose,200 mg/L boric acid,75 mg/L calcium chloride),among which the pollen germination rate of Lilium brownii var.viridulum Baker and‘Strawberry Event’was higher than that of other parent varieties,and the pollen vitality was better.The hybrid fruit expansion rate of distance hybridization between Lilium lancifolium Thunb.and Asiatic hybrids was higher,among which the hybridization with‘Strawberry Event’had the highest rate of fruit enlargement with 14.7%,and only this combination successfully obtained two hybrid offspring.【Conclusion】This study shows that the hybridization compatibility between Lilium lancifolium Thunb.and Asiatic lily hybrid line is better,which provides technical support for distant hybrid breeding in the future.
