摘要
目的:应用数据挖掘和网络药理学的研究方法,探讨从“瘀毒”论治冠心病的古方中药配伍规律并挖掘其核心中药组,探索核心中药组的作用机制。方法:利用古今医案云平台对从数据库中提取的古方进行数据挖掘,提取核心中药组,应用网络药理学方法探究其治疗冠心病的作用机制。结果:提取得到古方208首。其中,频次排序居前3位的中药为当归、川芎、人参,四气包含温、平、微寒、寒、微温等,五味以辛、苦、甘为主,归经以肝、脾经为主,得到的核心中药组为人参、当归、川芎。核心中药组共有中药活性成分28个,中药活性成分靶点685个,疾病靶点1450个,共同靶点190个。核心靶点为白细胞介素6、肿瘤坏死因子、蛋白激酶B1和过氧化物酶体增殖物激活受体γ等40个,当归-人参-川芎核心中药组治疗冠心病是通过多通路来发挥作用的。结论:从“瘀毒”论治冠心病的古方通过多成分、多靶点、多通路发挥治疗冠心病的作用,其作用机制与现代研究中的“瘀毒理论”“炎症反应学说”具有相通之处,可为今后的相关研究提供可靠的理论基础。
OBJECTIVE:According to research methods of data mining and network pharmacology,to probe into the medication rules and mechanism of ancient traditional Chinese medicine prescription in the treatment of coronary heart disease from the theory of“Stasis Toxin”,and to explore its core traditional Chinese medicine group and mechanism of action.METHODS:The ancient prescriptions extracted from the literature database were data-mined by using the Cloud Platform of Ancient and Modern Medical Cases,and the core group of traditional Chinese medicines was extracted.The mechanism in the treatment of coronary heart disease was investigated by network pharmacology.RESULTS:Totally 208 ancient prescriptions were extracted.Among them,the top three ranked by frequency of traditional Chinese medicines were Angelicae sinensis radix,Chuanxiong rhizoma,and Panax ginseng C.A.Mey.The four Qi contained warmth,calmness,slight coldness,coldness,and slight warmth.The five flavors were mainly pungent,bitter,and sweet,with the liver meridian and the spleen meridian as the main meridian.The core group of traditional Chinese medicine was Panax ginseng C.A.Mey,Angelicae sinensis radix,and Chuanxiong rhizoma.There were 28 traditional Chinese medicine active components,685 traditional Chinese medicine active component targets,1450 disease targets and 190 common targets in the core group of traditional Chinese medicine.There were 40 core targets,including interleukin 6,tumor necrosis factor,protein kinase B1 and PPARG.Angelicae sinensis radix-Panax ginseng C.A.Mey-Chuanxiong rhizoma plays a role in the treatment of coronary heart disease through multiple pathways.CONCLUSIONS:Ancient prescription in the treatment of coronary heart disease from the theory of“Stasis Toxin”plays a role in the treatment of coronary heart disease through multi-components,multi-targets and multi-pathways,and its mechanism has similarities with the“Theory of Stasis Toxin”and“Theory of Inflammation”in modern research,which provides reliable theoretical basis for future related research.
