复元康胶囊对睡眠及惊厥影响实验研究

摘要 目的:研究复元康胶囊对阈剂量戊巴比妥钠所致小鼠睡眠时间及睡眠潜伏期的影响;研究复元康胶囊对戊四氮所致惊厥小鼠惊厥潜伏期和存活时间的影响。探讨复元康胶囊对中枢神经系统的影响。方法:将60只雌雄各半的KM小鼠分为2批,每批30只,每批随机分为空白组、复元康胶囊组和艾司唑仑组或苯妥英钠组,每组10只。除空白组给予蒸馏水外,其余各组均给予相同体积的受试药物,连续给药3 d后,各组末次给药1 h后腹腔注射戊巴比妥钠或戊四氮,观察记录小鼠翻正反射消失时间和恢复时间或小鼠惊厥潜伏期、存活时间。结果:(1)本实验艾司唑仑组与复元康胶囊组小鼠睡眠潜伏期时间少于空白组(P<0.05),睡眠持续时间显著多于空白组(P<0.05),艾司唑仑组小鼠睡眠潜伏期时间显著少于复元康胶囊组(P<0.01),小鼠睡眠持续时间显著多于复元康胶囊组(P<0.01);(2)苯妥英钠组和复元康胶囊组小鼠惊厥潜伏期时间与小鼠存活时间均多于空白组(P<0.01),苯妥英钠组小鼠惊厥潜伏期时间及小鼠存活时间显著多于复元康胶囊组(P<0.01)。结论:复元康胶囊对戊巴比妥钠阈下催眠具有一定的协同作用,对戊四氮所致小鼠惊厥有一定拮抗作用,提示复元康胶囊对中枢神经系统具有一定的镇静催眠、抗惊厥作用,推测复元康胶囊治疗精神分裂症阳性症状效果显著,其作用机制可能与镇静、抗惊厥作用有关。 Objective:To Study the effect of Fuyuankang capsule on sleep time and sleep latency of mice caused by threshold dose of sodium pentobarbital;to study the effect of Fuyuankang capsule on convulsive latency and survival time of mice with convulsions caused by pentylenetetrazol.To investigate the effect of Fuyuankang capsule on the central nervous system.Methods:Sixty male and female KM mice were divided into 2 batches of 30 mice each,and each batch was randomly divided into a blank group,a Fuyuankang capsule group,and an eszopiclone group or phenytoin sodium group,with 10 mice in each group.Except for the blank group,which was given distilled water,all the other groups were given the same volume of the test drug,and after 3 consecutive days of administration,sodium pentobarbital or pentylenetetrazol was injected intraperitoneally 1 hour after the last administration of the drug in each group,and the time of disappearance of the flip-flop reflex and the time of recovery,or the latent period of convulsions and the time of survival in mice were observed and recorded.Results:①In this experiment,the sleep latency of mice in the estazolam group and Fuyuankang capsule group was less than that in the blank group(P<0.05),and the sleep duration was significantly longer than that in the blank group(P<0.05).The sleep latency in estazolam group was significantly less than that in Fuyuankang capsule group(P<0.01),and the sleep duration in estazolam group was significantly longer than that in Fuyuankang capsule group(P<0.01).②The convulsion latency and survival time of mice in phenytoin sodium group and Fuyuankang capsule group were significantly longer than those in blank group(P<0.01).The convulsion latency and survival time in phenytoin sodium group were significantly longer than those in Fuyuankang capsule group(P<0.01).Conclusion:Fuyuankang capsule has a certain synergistic effect on subthreshold hypnosis of pentobarbital sodium and a certain antagonistic effect on convulsion induced by pentylenetetrazol in mice,suggesting that Fuyuankang capsule has certain sedative and hypnotic and anticonvulsant effects on the central nervous system.It is speculated that Fuyuankang capsule is effective in the treatment of positive symptoms of schizophrenia,and its mechanism may be related to sedation and anticonvulsant effects.
