摘要
目的:探讨肾脑复元汤联合人脐带间充质干细胞(hUC-MSCs)移植对脑缺血再灌注损伤模型大鼠神经细胞凋亡的影响。方法:将75只SD大鼠随机分成假手术组、模型组、肾脑复元汤组、hUC-MSCs组、联合组(肾脑复元汤联合hUC-MSCs移植),大脑中动脉栓塞(MCAO)法制备脑缺血再灌注损伤模型大鼠,每组又分为术后第3天、7天、14天三个时间,术前24h、术后2h及处死前1h对大鼠进行神经功能缺损评分,免疫组化和Western Blot检测脑组织B淋巴细胞瘤-2基因(Bcl-2)、Bcl-2关联X蛋白(Bax)蛋白表达情况。结果:与假手术组比较,模型组神经功能评分显著升高,Bcl-2蛋白表达降低,Bax蛋白表达升高,差异具有统计学意义(P<0.05,P<0.01);与模型组比较,肾脑复元汤组、hUC-MSCs组、联合组(术后第3、7、14天)神经功能评分均显著降低,Bcl-2蛋白表达增多,Bax蛋白表达减少,差异具有统计学意义(P<0.05,P<0.01);与肾脑复元汤组和hUC-MSCs组比较,联合组(术后第3、7、14天)神经功能评分显著降低,Bcl-2蛋白表达增多,Bax蛋白表达减少,差异具有统计学意义(P<0.05,P<0.01)。结论:肾脑复元汤联合hUC-MSCs移植可能通过抑制神经细胞凋亡来促进脑缺血再灌注损伤模型大鼠神经功能修复。
Objective:Explore the impact of Shennao Fuyuan Decoction combined with human umbilical cord mesenchymal stem cells(hUC-MSCs)transplantation on neuronal apoptosis in a rat model of cerebral ischemia-reperfusion injury.Methods:75 Sprague-Dawley rats were randomly assigned to five groups:sham-operation group,model group,Shennao Fuyuan Decoction group,hUC-MSCs transplantation group,and the combination group(Shennao Fuyuan Decoction combined with hUC-MSCs transplantation).The MCAO method was used to create the rat model of cerebral ischemia-reperfusion injury.Each group was further divided into three time periods:the 3rd,7th,and 14th day after surgery.Neurological function deficits were assessed 24 hours before surgery,2 hours after surgery,and 1 hour before euthanasia.Immunohistochemistry and Western Blot techniques were employed to measure the expression of B-cell lymphoma-2 gene(Bcl-2)and Bcl-2 associated X protein(Bax)in brain tissue.Results:In comparison to the sham-operation group,the model group exhibited a statistically significant elevation in neurological function score,a reduction in Bcl-2 protein expression,and an increase in Bax protein expression(P<0.05,P<0.01);Conversely,when compared to the model group,the Shennao Fuyuan Decoction group,hUC-MSCs group,and combination group(on postoperative days 3,7,and 14)all demonstrated a significant decline in neurological function scores,an increase in Bcl-2 protein expression,and a decrease in Bax protein expression(P<0.05,P<0.01);In comparison to the Shennao Fuyuan Decoction group and hUC-MSCs group,the combined group exhibited a statistically significant reduction in neurological function scores,an elevation in Bcl-2 protein expression,and a decline in Bax protein expression on postoperative days 3,7,and 14(P<0.05,P<0.01).Conclusion:The co-administration of Shennao Fuyuan Decoction and hUC-MSCs transplantation has the potential to facilitate neural function restoration in rats with cerebral ischemia-reperfusion injury through the inhibition of neuronal apoptosis.
