经典名方保元汤提取工艺优化比较

Comparative on the Optimization of Extraction Process ofClassical Prescription Baoyuan Decoction

目的:优选保元汤的煎煮和回流提取工艺,为经典名方复方制剂开发利用提供参考。方法:按照《古代经典名方关键信息表(25首方剂)》中考证的保元汤现代对应情况,以煎煮时间、提取次数、加水体积为考察因素,以出膏率、人参皂苷Rb1、Rg1、Re含量为考察指标,采用L_(9)(3^(4))正交试验法筛选出保元汤传统煎煮法及回流提取法最佳提取工艺,并对两种提取方法进行比较。结果:保元汤最佳煎煮工艺为12倍水量,煎煮时间1 h并煎煮3次,回流提取最佳工艺是12倍水量2 h并回流3次。分别在最佳提取条件下,回流提取法比煎煮提取法所得的保元汤中三种人参皂苷Rb1、Rg1、Re含量及出膏率更高。结论:经典名方保元汤可采用回流提取法生产,采用12倍水量2 h并回流3次工艺稳定可行,可为其复方制剂开发和应用提供依据,同时为经典名方制剂研究提供思路。

Objective:To optimize the decoction and reflux extraction process of Baoyuan Decoction and provide reference for the development and utilization of Classical Prescription.Methods:According to the modern correspondence of Baoyuan Decoction verified in the Key Information Table of Ancient Classic Famous Prescriptions(25 Prescriptions),decoction time,extraction frequency,and added water volume were taken as the examination factors,and the extract rate,ginsenoside Rb1,and Rg1,Re content were taken as the examination indicators,and the L_(9)(3^(4))orthogonal experiment method was used to screen the optimal extraction process for Baoyuan decoction using traditional decoction method and reflux extraction method,and the two extraction methods were compared.Results:The optimal decoction process of Baoyuan decoction was 12 times the amount of added water for 1h and decocting 3 times.The optimal reflux extraction process was 12 times the amount of added water for 2h and refluxing three times.Under the optimal extraction conditions,the reflux extraction method resulted in higher content of the three ginsenosides Rb1,Rg1,Re,and higher extraction rate in the Baoyuan Decoction compared to the decoction extraction method.Conclusion:The classic prescription Baoyuan Decoction can be produced by the reflux extraction method.The process of using 12 times the amount of added water for 2h and refluxing three times is stable and feasible,which can provide a basis for the development and application of its compound preparations and provide ideas for the research on classic prescription preparatio.
