摘要
为了深入研究MYB3R基因在多刺绿绒蒿逆境适应和次生代谢物积累中的作用机制,本研究基于多刺绿绒蒿全长转录组测序数据,鉴定和克隆了多刺绿绒蒿一个MYB3R基因,并进行生物信息学分析。结果表明,MYB3R基因的完整开放阅读框(ORF)长1590 bp,编码529个氨基酸,具有保守结构域;该蛋白无跨膜结构域,为亲水性、不稳定蛋白,且为非分泌蛋白;二级结构中的α-螺旋、β-转角、无规卷曲、氨基酸残基构成的延伸链分别为28.54%、3.21%、64.84%和3.40%;建模的MhMYB3R蛋白质主要由α-螺旋组成,形成了一个相对对称的立体结构;系统进化树的结果表明多刺绿绒蒿MYB3R蛋白氨基酸序列和罂粟有高达97%的相似度。MYB3R基因的成功克隆和预测结果为深入研究MYB转录因子在多刺绿绒蒿生长发育、次生代谢产物、逆境适应等方面提供参考。
In order to deeply study the mechanism of MYB3R gene in stress adaptation and secondary metabolite accumulation of Meconopsis horridula,this study cloned a MYB3R gene of Meconopsis spinosa by polymerase chain reaction(PCR)based on the annotation information of MYB gene in transcriptome,and analyzed it by bioinformatics method.The results showed that the complete open reading frame(ORF)of MYB3R gene was 1590 bp,encoding 529 amino acids with conserved domain.The transmembrane domain of the protein was unstable,hydrophilic and non-secretory.In the secondary structure,α-helix was 28.54%,β-turn was 3.21%,random curl was 64.84%,and extension chain composed of amino acid residues was 3.40%.The modeled MhMYB3R protein was mainly composed ofα-helix,forming a relatively symmetrical three-dimensional structure.The results of phylogenetic tree showed that the amino acid sequence of MYB3R protein of Meconopsis horridula was 97%similar to that of Papaver somniferum.The successful cloning and prediction results of MYB3R gene provide theoretical basis for further study of MYB transcription factors in the growth and development,secondary metabolites and adaptation to adversity of Meconopsis horridula.
