摘要
采用絮凝法对马铃薯淀粉废水中的马铃薯蛋白质进行回收,以蛋白质回收率为评价指标,采用响应面设计对回收工艺条件进行优化。结果表明,最佳回收工艺条件是壳聚糖用量6.09 mg/100 mL废水,pH值4.06,温度41.22℃,处理时间39.35 min,在此条件下的蛋白质回收率为77.8645%。絮凝后的粗蛋白经AKTAgo蛋白纯化仪的DEAE GE Healthcare色谱柱分离纯化的马铃薯蛋白的热变性温度为51.2℃。研究表明壳聚糖絮凝法可较好地回收马铃薯废水蛋白。
Potato protein was recovered from potato starch wastewater by flocculation method.Response surface design was used to optimize the recovery process with protein recovery as evaluation index.The results showed that the optimal recovery conditions were as follows:Chitosan dosage was 6.09 mg/100 mL wastewater,pH value was 4.06,temperature was 41.22℃,treatment time was 39.35 min,and protein recovery was 77.8645%under these conditions.The thermal denaturation temperature of the purified potato protein was 51.2℃by DEAE GE Healthcare column of AKTAgo protein purification instrument.The results showed that chitosan flocculation could recover protein from potato wastewater.
