摘要
目的研究新疆阿魏含药血清对HeLa细胞凋亡及相关蛋白Survivin和Caspase-3的影响。方法大鼠灌胃新疆阿魏获得含药血清,用含药血清处理HeLa细胞,采用CCK-8法测定细胞增殖抑制率,采用Hoechst33258染色联合流式细胞仪检测细胞凋亡,运用Western Blot法检测Survivin和Caspase-3蛋白表达。结果各浓度含药血清对细胞增殖抑制率分别为(11.4%±2.3%)、(14.9%±3.1%)、(25.3%±3.2%),细胞凋亡率分别为(29.4%±9.9%)、(35.8%±5.3%)、(43.1%±4.7%),各浓度含药血清组细胞中Survivin蛋白降低、Caspase-3蛋白升高(P<0.05)。与空白对照组比较,上述结果差异有统计学意义(P<0.01)。结论新疆阿魏含药血清促进HeLa细胞凋亡,可能与抑制Survivin蛋白表达和提高Caspase-3蛋白表达有关。
Objective To explore the impact of serum containing Ferula sinkiangensis from Xinjiang on the apoptosis of HeLa cells by assessing the expression levels of Survivin and Caspase-3 proteins,which are associated with the regulation of apoptosis.Methods Serum containing Ferula sinkiangensis was obtained by administering the substance to rats via gavage,and the serum was then used to treat HeLa cells.The Cell Counting Kit-8(CCK-8)method was used to determine the inhibition rate of cell proliferation.Apoptosis was evaluated using flow cytometry and Hoechst 33258 staining.The expressions of Survivin and Caspase-3 proteins were detected using Western Blot analysis.Results The proliferation inhibitory rates of the drugcontaining serum at various concentrations were 11.4%±2.3%,14.9%±3.1%,and 25.3%±3.2%,respectively.The apoptosis rates in the treated groups were 29.4%±9.9%,35.8%±5.3%,and 43.1%±4.7%,respectively.In all treated groups,there was a decrease in Survivin protein and an increase in Caspase-3 protein.As above,these differences were statistically significant compared to the control group(P<0.01).Conclusion Serum containing Ferula sinkiangensis from Xinjiang can promote apoptosis in HeLa cells,which may be related to the suppression of Survivin protein and the upregulation of Caspase-3 protein expression.
