摘要
【目的】挖掘与燕麦倒伏性状相关的SRAP分子标记,为燕麦倒伏相关性状的研究及分子标记辅助育种提供遗传基础。【方法】以62份燕麦种质为试材,测定其表型性状、茎秆力学特性以及茎秆生理生化指标,并进行描述性和相关性分析;利用筛选的10对SRAP引物组合对其进行多态性和关联分析。【结果】供试燕麦种质的茎壁厚度、茎秆弯曲强度、茎秆穿刺强度、茎秆压碎强度、倒伏指数、产量,以及茎秆第2节间的可溶性糖含量、纤维素含量、氮含量、磷含量和钾含量等11个性状差异较大;燕麦的株高、重心高度、穗位高和茎秆第2节间氮含量与倒伏指数呈极显著正相关,而茎秆第2节间的可溶性糖、纤维素、木质素和钾含量及茎秆穿刺强度与倒伏指数呈显著负相关。SRAP多样性分析显示,多态信息含量均值为0.4076,遗传多样性较高。关联分析分别检测到46个和25个标记位点与倒伏相关性状关联,其中位点A17B2和T4G4与多个倒伏性状关联。【结论】供试燕麦种质遗传差异大,多样性指数较高。关联分析找到与燕麦倒伏相关性状显著关联的SRAP分子标记,可用于燕麦分子标记辅助育种。
【Objective】The study mined SRAP molecular markers related to oat collapse traits to provide genetic basis for studying oat collapse-related traits and molecular marker-assisted breeding.【Method】A total of 62 oat germplasm samples were used to determine their phenotypic characters,stem mechanical properties and physiological and biochemical indexes,and to conduct descriptive and correlation analysis.The polymorphism and association of 10 pairs of SRAP primers were analyzed.【Result】The stem wall thickness,stem bending strength,stem penetration strength,stem crushing strength,lodging index,yield,soluble sugar content,cellulose content,nitrogen content,phosphorus content and potassium content in the second stem node of oat germplasm were significantly different.The plant height,center of gravity height,ear height and nitrogen content in stem second node were positively correlated with lodging index,while contents of soluble sugar,cellulose,lignin and potassium in second stem node and the intensity of stem insertion were negatively correlated with lodging index.SRAP diversity analysis showed tha mean polymorphic information content was 0.4076,indicating high genetic diversity.Association analysis detected 46 and 25 loci associated with lodging traits,respectively,and loci A17B2 and T4G4 were associated with multiple lodging traits.【Conclusion】Oat germplasm had great genetic difference and high diversity index.Correlation analysis found that SRAP molecular markers were significantly associated with lodging related traits in oat,which could be used for marker assisted breeding in oat.
