目的综述白介素-6(IL-6)和肝细胞癌的关系,总结IL-6在肝细胞癌治疗中的最新研究进展以及存在的潜在问题。方法应用中国知网及PubMed检索系统,以“白介素-6、肝细胞癌、肿瘤治疗、潜在问题”为中文关键词,以“IL-6、hepatocellular carcinoma,oncotherapy,potential issues”为英文关键词,检索2014-01-01-2024-04-30发表的相关文献。共检索到中文文献89条,英文文献603条。纳入标准:(1)IL-6定义及相关信号通路;(2)IL-6在肝细胞癌中的相关研究;(3)IL-6在肝细胞癌治疗中的应用;(4)IL-6治疗面临的潜在问题。排除标准:(1)数据陈旧文献;(2)会议和评论类等文章;(3)内容相似或重复的文献。最终共纳入文献59篇。结果IL-6是一种多功能炎性细胞因子,主要通过IL-6/信号转导与转录激活因子3(STAT3)信号通路发挥生物学效应。IL-6在肝癌患者中表达增加,异常激活的IL-6/STAT3信号通路可通过影响肿瘤细胞增殖、迁移、侵袭、血管生成和凋亡等过程参与肝细胞癌发生发展,且与预后密切相关。通过探索IL-6与肝细胞癌的相互作用,概括基于IL-6相关治疗方式,包括抗体药物治疗、小分子抑制剂治疗、基因转染、基因编辑技术以及综合治疗策略。揭示IL-6相关治疗药物,为肝细胞癌患者提供更有效的治疗选择,改善患者的生活质量和预后。结论IL-6在肝细胞癌的发生、增殖、侵袭及转移中起着重要作用,基于IL-6相关的治疗方式为肝细胞癌的治疗提供新的思路,但仍面临着诸多挑战。

Objective To introduce the relationship between Interleukin-6(IL-6)and hepatocellular carcinoma,and to summarize the latest research progress on IL-6 in the treatment of hepatocellular carcinoma and potential issues.Methods China National Knowledge Infrastructure and PubMed databases were used to find relevant literature published from January 1,2014 to April 30,2024,with"IL-6,hepatocellular carcinoma,oncotherapy,potential issues"as the Chinese and English keywords.A total of 89 Chinese articles and 603 English articles were retrieved.Inclusion criteria:(1)The definition of IL-6 and its related signaling pathways;(2)Studies on IL-6 on hepatocellular carcinoma;(3)Applications of IL-6 in the treatment of hepatocellular carcinoma;(4)Potential issues in the treatment of IL-6.Exclusion criteria:(1)Outdated literature;(2)Conference and commentary articles;(3)Literature with similar or duplicate content.A total of 59articles were included.Results IL-6 is a multifunctional inflammatory cytokine that exerts its biological effects primarily through the IL-6/STAT3 signaling pathway.IL-6 is up-regulated in hepatocellular carcinoma patients,and the aberrant activation of the IL-6/STAT3 signaling pathway can contribute to hepatocellular carcinoma progression by affecting tumor cell proliferation,migration,invasion,angiogenesis,and apoptosis,and is closely related to prognosis.By exploring the interaction between IL-6 and hepatocellular carcinoma,this article summarizes IL-6 related treatment methods,including antibody drug therapy,small molecule inhibitor therapy,gene transfection,gene editing technology,and comprehensive treatment strategies.Revealing IL-6-related therapeutic drugs provides more effective treatment options for hepatocellular carcinoma patients,and improves their quality of life and prognosis in the future.Conclusions IL-6 plays a crucial role in the occurrence,proliferation,invasion,and metastasis of hepatocellular carcinoma.Therapeutic approaches based on IL-6 provide new insights into hepatocellular carcinoma treatment,yet still face numerous challenges.
