摘要
目的本研究旨在探索乳腺癌患者中重度抑郁状态下长链非编码RNA(lncRNA)的表达变化,并评估这些lncRNA在抑郁症筛查中的潜在诊断价值。方法研究对象为在某医院接受治疗且诊断后生存五年及以上的乳腺癌患者,分为中重度抑郁组(20例,PHQ-9≥15)和无抑郁组(20例,PHQ-9=0~4)。通过高通量测序技术对全血浆lncRNA进行测序,筛选差异表达显著的lncRNA,并利用定量PCR进行验证。通过受试者工作特征(ROC)曲线分析评估lncRNA在抑郁症筛查中的诊断效能。结果结果显示,共有173个lncRNA在中重度抑郁组和无抑郁组之间存在显著差异表达。特别是Dr1、Gm2694和Nr2f2在中重度抑郁组中显著上调,分别具有3.00、3.01和2.58的Fold Change(P<0.005)。ROC曲线分析结果表明,Dr1、Gm2694和Nr2f2的曲线下面积(AUC)分别为0.85、0.83和0.80,联合使用Dr1和Gm2694时,AUC达到0.88。结论本研究揭示了乳腺癌患者中重度抑郁状态下lncRNA表达谱的显著差异,并初步证实了这些lncRNA在抑郁症筛查中的潜在诊断价值。这些发现为乳腺癌患者的心理健康管理提供了新的理论依据,也为未来开发新型的抑郁症生物标志物奠定了基础。
Objective This study aims to explore the expression changes of long non-coding RNA(lncRNA)in breast cancer patients with moderate to severe depression,and to evaluate the potential diagnostic value of these lncRNAs in depression screening.Methods The study subjects were breast cancer patients who had been treated in a hospital and survived for five years or more after diagnosis.They were divided into a moderate-to-severe depression group of 20 patients with a PHQ-9 score of 15 or higher and a non-depressed group of 20 patients with a PHQ-9 score of 0 to 4.The whole plasma lncRNA was sequenced by high-throughput sequencing technology to screen significantly differentially expressed lncRNA,and verified by quantitative PCR.The diagnostic efficacy of lncRNA in depression screening was evaluated by receiver operating characteristic(ROC)curve analysis.Results The results showed that a total of 173 lncRNAs had significantly different expression between the moderate-to-severe depression group and the non-depression group.In particular,Dr1,Gm2694,and Nr2f2 were significantly up-regulated in the moderate-to-severe depression group,with Fold Change P<0.005 of 3.00,3.01,and 2.58,respectively.The ROC curve analysis results showed that the area under the curve(AUC)of Dr1,Gm2694,and Nr2f2 were 0.85,0.83,and 0.80,respectively.When Dr1 and Gm2694 were used in combination,the AUC reached 0.88.Conclusions This study reveals significant differences in lncRNA expression profiles in moderate to severe depression in breast cancer patients,and preliminarily confirms the potential diagnostic value of these lncRNAs in depression screening.These findings provide a new theoretical basis for the management of mental health in breast cancer patients,and also lay the foundation for the development of new biomarkers for depression in the future.
