蚕丝蛋白的体外自组装动力学行为研究

摘要 丝素蛋白具有优异的力学性能和机械强度,丝素蛋白自组装成纤维可以进一步增强这种性能。以家蚕蚕茧为原料,首先提取制备再生丝素蛋白,采用圆二色谱对提取丝素进行结构分析和结构鉴定。在此基础上,进一步采用浊度实验、热稳定性实验研究提取的丝素蛋白分子纤维化能力、热稳定性,结果表明,丝素蛋白自组装的最佳条件为:浓度4 mg/mL在70℃组装6 h;并且所提取的丝素蛋白组装前和组装后变性温度分别为90.36℃和114.52℃,组装后热稳定性提高;旋转流变实验结果表明丝素蛋白所形成的凝胶网络结构稳定,行为类似于固体;细胞生物学实验表明,再生丝素蛋白溶液利于细胞增殖。 Silk fibroin has excellent mechanical properties and mechanical strength.Self-assembly of silk fibroin into fibers can further enhance the properties.In this paper,silk fibroin protein was extracted from silkworm cocoon,and its structure was analyzed and identified by circular dichroism spectrum.On this basis,the turbidity and thermal stability were used to study the fibrotic ability and thermal stability of silk fibroin,the optimal conditions for silk fibroin self-assembly were as follows:4 mg/mL of silk fibroin was assembled at 70℃for 6 h,and the denaturation temperatures of the extracted silk fibroin before and after assembly were 90.36℃and 114.52℃,respectively,and the thermal stability of the extracted silk fibroin was improved after assembly The results of rotational rheological experiments show that the gel network formed by silk fibroin is stable and behaves like a solid.Cell biology experiments showed that silk fibroin solution is more conducive to cell value-added.
