摘要
维生素B_(1)(VB_(1))是临床治疗中常用的B族维生素,对于人体多种疾病的治疗起到重要作用。利用单链A30-DNA为模板合成的荧光金纳米簇(AuCNs)作为荧光探针,构建了一种快速免标记荧光传感方法来检测VB_(1)。由于VB_(1)与金纳米簇的表面基团的相互作用,它能够有效地猝灭金纳米簇的荧光。在优化条件下,对不同VB_(1)浓度进行了检测,线性方程为:F 0-F=18.034+437.16 C VB_(1)(C VB_(1)为VB_(1)浓度,μmol/L),检测线性范围为0.01~1μmol/L,检测限为3 nmol/L。在实际样品的检测中,回收率在97.5%~106.3%之间,相对标准偏差(RSD)在1.5%~2.3%之间。该方法不需要设计复杂的DNA序列,也不需要复杂的荧光标记操作,实现了较广浓度范围内硫胺素药物的检测,具有简便、高灵敏度和选择性好等优点。
Vitamin B_(1)(VB_(1))is a thiamine drug commonly used in clinical treatment and has been clinically proved effective in treating specific human diseases.A rapid and label-free fluorescence sensing method has been developed for VB_(1)detection in pharmaceutical sample by using A30-ssDNA-templated synthesis of gold nanoclusters(AuNCs)as fluorescent probe.Due to the interaction between VB_(1)and A30-ssDNA-templated AuNCs,VB_(1)can effectively quench the fluorescence of A30-ssDNA-templated AuNCs.Under optimized conditions,a series concentration of VB_(1)were detected.The linear equation was F 0-F=18.034+437.16 C VB_(1),The relative fluorescence intensity displayed an good linear relationship with VB_(1)concentration in the range from 0.01 to 1μmol/L and a detection limit of 3 nmol/L.In pharmaceutical sample detection,the recoveries ranged from 97.5%to 106.3%,and the relative standard deviation ranged from 1.5%to 2.3%.This method is free of complex DNA sequence design or any fluorescence dye label.It realizes the detection of thiamine drugs in a wide range of concentrations,and has the advantages of simplicity,high sensitivity and good selectivity.
