丁香多糖的提取及其对嗜水气单胞菌抑制活性研究

摘要 本文通过单因素实验,考察了提取温度、提取时间、料液比和醇沉体积分数对丁香多糖提取率的影响,并通过正交试验,以多糖提取率作为评价指标,进一步优化丁香多糖提取工艺。采用滤纸片扩散法考察了丁香多糖对嗜水气单胞菌抑制活性。研究发现,该方法在实验时间内稳定,精密度、重复性、稳定性良好。加样回收试验表明,该方法平均回收率为99.82%,RSD为1.23%。深入研究发现,将提取温度设定为90℃,提取时间设定为120 min,料液比设定为1∶60,醇沉体积分数设定为80%,丁香多糖提取工艺最佳。进一步研究发现,丁香多糖对嗜水气单胞菌表现出较好的抑制效果,MIC为12.5 mg/mL。该工艺简单稳定,丁香多糖抑制嗜水气单胞菌明显,为该资源的进一步开发提供了研究基础。 This article investigates the effects of extraction temperature,extraction time,solid-liquid ratio,and alcohol precipitation volume fraction on the extraction rate of syzygium aromaticum polysaccharides through single factor experiments.Through orthogonal experiments,the polysaccharide extraction rate is used as an evaluation index to further optimize the syzygium aromaticum polysaccharide extraction process.The inhibitory activity of syzygium aromaticum polysaccharides on aeromonas hydrophila was investigated using filter paper diffusion method.Research has found that this method is stable within the experimental time,with good precision,repeatability,and stability.The sample recovery test showed that the average recovery rate of this method was 99.82%,with an RSD of 1.23%.Through in-depth research,it was found that setting the extraction temperature to 90℃,extraction time to 120 min,solid-liquid ratio to 1∶60,and alcohol precipitation volume fraction to 80%resulted in the optimal extraction process for syzygium aromaticum polysaccharides.Further research has found that syzygium aromaticum polysaccharides exhibit good inhibitory effects on aeromonas hydrophila,with a MIC of 12.5 mg/mL.This process is simple and stable,and the polysaccharides from syzygium aromaticum significantly inhibit aeromonas hydrophila,providing a research basis for further development of this resource.
