摘要
目的 采用双样本孟德尔随机化方法探讨多囊卵巢综合征(PCOS)与多发性骨髓瘤(MM)的因果关系.方法 以全基因组关联分析(GWAS)数据为基础,使用MR分析探讨PCOS与MM之间的因果关联.筛选与PCOS密切关联的单核苷酸多态性(SNPs),使用逆方差加权(IVW)方法进行分析,使用优势比(OR值)进行因果关系评估.采用IVW和MR-Egger进法Cochran Q异质性检验,用MR_pleiotropy_test函数、MR PRESSO检验多效性,MR_leaveoneout_plot函数分析敏感性.结果 IVW分析结果显示,在PCOS人群中多发性骨髓瘤患病风险增加(OR:1.415,95%CI:1.046-1.916,P=0.024),与其他四种分析方法结果一致.从森林图和散点图可以直观看出,PCOS与MM的关系呈正相关.反向MR分析时则未能筛选出足够的SNP.结论 PCOS患者发生MM的风险增高,提示PCOS患者应提高对MM疾病的认知,加强防范措施.
Objective The objective of this research was to assess the potential causal relationship between Polycystic Ovary Syndrome(PCOS)and Multiple Myeloma(MM).Methods A two-sample Mendelian randomization(MR)analysis was performed using data from a genome-wide association study(GWAS),to explore the potential causal relationship between PCOS and MM.Single nucleotide polymorphisms(SNPs)linked to PCOS were selected.The inverse variance weighting(IVW)method was used as the analytical approach and odds ratio(OR)was employed to investigate the causal relationship between PCOS and MM.To assess the robustness of the analysis results,sensitivity analyses were performed with incorporating tests for heterogeneity,pleiotropy,and the leave-one-out method.Results The IVW method indicated a heightened risk of MM among PCOS patients(OR:1.415,95%CI:1.046-1.916,P=0.024).Similar results were obtained through analyses utilizing four other methods.Forest plots and scatter plots demonstrated a positive correlation between PCOS and MM.The reverse MR analysis did not successfully filter out a sufficient number of SNPs.Conclusion A causal relationship exists between PCOS and MM,indicating that PCOS patients should be vigilant of the potential risk of MM.It is suggested that PCOS patients should pay close attention to the risk of MM,strengthen the preventive measures.
