组织RP11-757G1.5水平对胃癌根治术患者预后的评估价值

Prognostic value of tissue RP11-757G1.5 level in patients with gastric cancer undergoing radical gastrectomy

目的探讨胃癌组织中的长链非编码RNA RP11-757G1.5水平及其预后评估价值。方法采用实时荧光定量PCR检测RP11-757G1.5在98对胃癌组织及对应癌旁组织中的表达差异,基于临床病理资料和随访数据分析RP11-757G1.5的临床价值。结果胃癌组织的RP11-757G1.5水平高于癌旁组织(4.424±0.173 vs.1.686±0.076,t=14.508,P<0.001)。RP11-757G1.5在胃癌组织中表达显著与TNM分期(χ^(2)=7.721,P=0.006)、分化程度(χ^(2)=8.279,P=0.004)、T分期(χ^(2)=5.528,P=0.019)和癌胚抗原状态(χ^(2)=4.125,P=0.042)有关。RP11-757G1.5低表达者的中位无病生存期未达,优于高表达者的38个月(HR=2.714,95%CI:1.613~4.743,P=0.008),经多因素分析证实RP11-757G1.5高表达是胃癌预后不良的危险因素(HR=2.438,95%CI:1.307~4.546,P=0.016)。结论RP11-757G1.5表达上调,是胃癌患者预后不良的预测因子,可能是一个有前途的胃癌生物标志物和治疗靶点。

Objective To investigate the level of long non-coding RNA RP11-757G1.5 in gastric cancer tissues and its prognostic value.Methods Ninety-eight pairs of gastric cancer tissues and corresponding adjacent tissues were collected.Differential expression of RP11-757G1.5 between groups were assessed via real-time quantitative PCR.The clinical value of RP11-757G1.5 was analyzed based on clinical pathological data and follow-up data.Results The RP11-757G1.5 level in gastric cancer tissues was higher than that in adjacent tissues(4.424±0.173 vs.1.686±0.076,t=14.508,P<0.001).RP11-757G1.5 level in gastric cancer tissues was associated with TNM staging(χ^(2)=7.721,P=0.006),differentiation degree(χ^(2)=8.279,P=0.004),T staging(χ^(2)=5.528,P=0.019),and carcinoembryonic antigen status(χ^(2)=4.125,P=0.042).The median disease-free survival of individuals with low RP11-757G1.5 expression was not reached,better than 38 months of individuals with high expression(HR=2.714,95%CI:1.613-4.743,P=0.008).High RP11-757G1.5 expression was a risk factor for poor prognosis in gastric cancer(HR=2.438,95%CI:1.307-4.546,P=0.016).Conclusion Upregulation of RP11-757G1.5 expression is a predictive factor for poor prognosis in gastric cancer patients and may be a promising biomarker and therapeutic target for gastric cancer.
