摘要
文章分别对采集自辽宁省鞍山市、云南省昆明市以及云南省贡山县的野生黑松露药材进行了加热回流法提取以及醇沉法制备黑松露提取物,经冷冻干燥法,制得了多组分的黑松露提取物,以苯酚-硫酸法对不同产地及不同处理方式下的黑松露提取物中的多糖含量进行研究,同时建立液相色谱-串联质谱法(LC-MS/MS)方法,测定不同产地样品提取液中L-麦角硫因含量。根据水提多糖含量检测结果,辽宁省鞍山(346.40±0.51 mg/g)>云南昆明(316.52±51.93 mg/g)>云南贡山(236.75±4.59 mg/g)。黑松露水提醇沉物,多糖云南贡山(611.84±4.43 mg/g)>云南昆明(420.31±3.45 mg/g)>辽宁鞍山(351.36±5.32 mg/g)。L-麦角硫因含量测定结果,云南省昆明(20.62±0.86 ng/g)>云南贡山县(26.03±1.71 ng/g)>辽宁鞍山(19.51±0.44 ng/g)。不同产地的野生黑松露样品间的黑松露总多糖含量差异较大,其麦角硫因含量也有明显区别。文章通过对不同产地黑松露样品中的多糖和麦角硫因进行含量检测,确定了不同产地的野生黑松露样品两种有效物质差距,对野生黑松露的最优产地进行了筛选,为黑松露在化妆品、医学、食品等领域的进一步开发和应用提供相应的理论基础。
In this paper,the wild black truffle extracts were collected from Anshan City,Liaoning Province,Kunming City,Yunnan Province and Gongshan County,Yunnan Province,which were extracted by heating reflux method and alcohol precipitation method,the multi-component black truffle extract was prepared by freeze-drying method,the polysaccharide content in the black truffle extract under different origins and different treatment methods was studied by phenol-sulfuric acid method,and the liquid chromatography-tandem mass spectrometry(LC-MS/MS)method was established.The content of L-ergothioneine in the extracts of samples from different origins was determined.According to the results of polysaccharide content extracted from water,the order was Anshan(346.40±0.51 mg/g)in Liaoning Province>Kunming in Yunnan Province(316.52±51.93 mg/g)>Gongshan in Yunnan Province(236.75±4.59 mg/g).According to the results of black truffle water extraction alcohol precipitate polysaccharides,the order was Gongshan in Yunnan(611.84±4.43 mg/g),>Kunming in Yunnan(420.31±3.45 mg/g),>Anshan in Liaoning(351.36±5.32 mg/g).The results of L-ergothioneine content were determined,and the order was Kunming,Yunnan Province(20.62±0.86 ng/g)>Gongshan County,Yunnan Province(26.03±1.71 ng/g)>Anshan,Liaoning(19.51±0.44 ng/g).The total polysaccharide content of black truffle was quite different among the wild black truffle samples from different origins,and the ergothioneine content was also significantly different.In this paper,the content of polysaccharides and ergothioneine in black truffle samples from different origins was detected,the gap between the two effective substances in wild black truffle samples from different origins was determined,and the optimal production areas of wild black truffle were screened,which provided a corresponding theoretical basis for the further development and application of black truffle in cosmetics,medicine,food and other fields.
