摘要
目的:探究五指毛桃总黄酮的提取工艺,比较不同产地五指毛桃的抗氧化活性。方法:采用Box-Behnken响应面法对五指毛桃总黄酮提取工艺进行优化,并测定不同产地五指毛桃总黄酮含量及抗氧化活性。结果:影响五指毛桃总黄酮提取效果的顺序为提取次数>提取时间>乙醇体积分数>液料比。最佳提取工艺为提取时间2.1 h、提取次数2次、乙醇体积分数70%、液料比20∶1,最佳提取工艺稳定、高效、可行,综合评价值为5.31 mg/g。广西玉林、广东河源、福建龙海、福建平和、福建南靖5个产地五指毛桃总黄酮含量分别为6.73 mg/g、5.28 mg/g、6.89 mg/g、7.96 mg/g和6.67 mg/g,有较强的抗氧化能力。不同产地五指毛桃总黄酮提取液对羟自由基清除率存在差异,由强到弱为广西玉林>福建平和>福建南靖>广东河源>福建龙海;不同产地五指毛桃提取液对DPPH自由基的半数清除浓度(IC50)为0.82~9.70 mg/ml,其中,福建南靖五指毛桃提取液对DPPH自由基的清除效果最好,而广西玉林五指毛桃提取液对DPPH自由基的清除效果最差;不同产地五指毛桃提取液对ABTS+自由基的清除能力存在差异,福建平和、广西玉林、福建龙海、广东河源、福建南靖五指毛桃提取液对ABTS+自由基的IC50分别为0.059 mg/ml、0.063 mg/ml、0.069 mg/ml、0.075 mg/ml、0.082 mg/ml。结论:本研究为不同产地五指毛桃的质量评价及五指毛桃后续的资源开发提供理论依据。
Objective:To explore the extraction process of general flavone of Wuzhimaotao and compare the antioxidant activity of Wuzhimaotao from different producing areas.Methods:Box-Behnken response surface method was used to optimize the extraction process of general flavone of Wuzhimaotao.The content and antioxidant activity of general flavone of Wuzhimaotao from different producing areas were determined.Results:The order affecting the extraction effect of general flavone of Wuzhimaotao was extraction times>extraction time>ethanol volume fraction>liquid-solid ratio.The optimum extraction process was as follows:extraction time was 2.1 h,extraction times was 2 times,ethanol volume fraction was 70%,and liquid-solid ratio was 20:1.The optimal extraction process was stable,efficient and feasible,and the overall evaluation value was 5.31 mg/g.The contents of general flavone of Wuzhimaotao from five producing areas,namely Yulin in Guangxi,Heyuan in Guangdong,Longhai in Fujian,Pinghe in Fujian and Nanjing in Fujian,were 6.73 mg/g,5.28 mg/g,6.89 mg/g,7.96 mg/g and 6.67 mg/g,respectively,showing strong antioxidant capacity.There were differences in the scavenging rate of hydroxyl radical of extracting solution of general flavone of Wuzhimaotao from different producing areas,and the order from strong to weak was Yulin in Guangxi>Pinghe in Fujian>Nanjing in Fujian>Heyuan in Guangdong>Longhai in Fujian.The half scavenging concentration(IC50)of DPPH free radical in the extracting solution in Wuzhimaotao from different producing areas was 0.82~9.70 mg/ml.Among them,the extracting solution in Wuzhimaotao from Nanjing in Fujian had the best scavenging effect on DPPH free radical,while the extracting solution in Wuzhimaotao from Yulin in Guangxi had the worst scavenging effect on DPPH free radical.There were differences in the scavenging ability of ABTS+free radicals in the extracting solution of Wuzhimaotao from different producing areas.The IC50 of ABTS+radicals of the extracting solution of Wuzhimaotao from Pinghe(Fujian),Yulin(Guangxi),Longhai(Fujian),Heyuan(Guangdong)and Nanjing(Fujian)were 0.059 mg/ml,0.063 mg/ml,0.069 mg/ml,0.075 mg/ml and 0.082 mg/ml,respectively.Conclusion:This study provides a theoretical basis for the quality evaluation and subsequent resource development of Wuzhimaotao from different producing areas.
