摘要
目的 观察激光损伤家兔视网膜后碱性成纤维细胞生长因子 (basicfibroblastgrowthfactor,bFGF)表达时相的变化 ,探讨bFGF在视网膜激光损伤后修复过程中的地位和作用。方法 用不同剂量的YAG倍频激光 (5 32nm)致伤有色家兔视网膜 ,于伤后 1、3、7、14d取眼底视网膜 脉络膜组织 ,提取细胞总RNA进行RNA斑点印迹 ,用地高辛标记的bFGFcDNA探针进行核酸杂交 ,检测bFGFmRNA的表达水平。结果 激光损伤后眼底组织中bFGFmRNA的表达水平明显提高 ,且随激光致伤量的加大 ,bFGFmRNA的表达水平亦呈进行性增加。结论 激光损伤后眼底组织中bFGFmRNA的表达明显增加 。
Objective To observe the expression of bFGF gene in rabbits' retinal post laser wound and its role of repair. Methods The eyes of pigmented rabbit were encleated on days 1,3,7,14 after the Qs Nd:YAG laser wound.RNA Dot Blotting was accessed the changes in the expression of bFGFmRNA with digoxigenin(DIG) labeled bFGF cDNA probes.Results Significantly increased expression of bFGFmRNA was found in the eyeground tissue and increasingly expression changed with the increase of laser wounded energy. Conclusions Significantly increased expression of bFGFmRNA suggests bFGF play an important role in wound healing process of rabbits retinal after laser wound.
