摘要 目的 :探讨自行设计的LHF(乳果糖 -组氨酸 -聚蔗糖 )器官液保存新生猪胰腺的效果。方法 :在 4± 1℃条件下 ,分别采用LHF液、UW液、Euro -Collins液 ,保存新生猪胰腺 12h、2 4h、36h后进行胰岛分离 ,观察保存前、后胰岛形态、功能、线粒体细胞色素氧化酶活力等变化。结果 :LHF液保存新生猪胰腺 36h后 ,组织结构基本完整 ,含水率低 ;胰岛分离后 ,超微结构正常 ,细胞存活率高 ,凋亡率低 ,分泌功能良好。细胞色素氧化酶活力正常 ,其指标与UW液类似 ,明显优于Euro -Collins保存液。结论 :LHF器官保存液可用作胰岛移植供体保存 ,具有较为广阔的应用前景。 Objective:To explore the cold preservation effects of self-designed LHF solution, composed of lactulose, histidine and Ficoll, on neonatal porcine pancreas. Methods:Islet isolation were performed after 12h, 24h and 36h in LHF, UW or Euro-Collins solution at 4±1℃. Morphological changes, functions and the activity of cytochrome oxidase were measured before and after islet isolation. Results:The amount of pancreatic tissue water content was not increased significantly when neonatal porcine pancreases were stored in LHF solution within 36 hours. Morphological changes of pancreas were slight. After isolation, the ultrastructure of islets was normal. Islet viability was 93.0%±3.4%,and apoptosis ratio was 3.7%±0.9%. Moreover, the stimulation index maintained a high level. The activity of cytochrome oxidase was not decreased significantly. The cold preservation effect of LHF solution on neonatal porcine pancreas was similar to that of UW solution, and much better than that of Euro-Collins solution. Conclusion:LHF solution may preserve neonatal porcine pancreas for 36 hours, and have a wide application prospect for islet transplantation.
