摘要
以牛血清白蛋白 戊二醛为交联剂 ,将乳酸氧化酶固定于鸡蛋膜上 ,氧电极作电化学敏感元件 ,制成乳酸氧化酶电极。传感器的线性响应范围为 5 .0×1 0 - 5~ 2 .5× 1 0 - 2 mol/L ,检测限为 2 .0× 1 0 - 5mol/L ,RSD为 1 .4% (n =1 1 ) ,回收率为 97.5 %~ 1 0 4 .0 %。该传感器可用于临床乳酸的测定。
Lactate oxidase was immobilized in egg membranes and crosslinked with bovine serum albumin with glutaraldehyde. The enzyme electrode was than constructed by putting the enzyme membrane onto an oxygen electrode. In phosphate buffers at pH 6.8 and at the temperature of 25~37 ℃, the response of the biosensor had a linear relationship with lactate concentration in the range of 5.0×10 -5 ~2 5×10 -2 mol/L with detection limit of 2.0×10 -5 mol/L and RSD of 1.4%( n =11). The biosensor was used for clinical assay of lactate in plasma and other body fluids.
