摘要 目的 探讨供体脾脏中的T细胞在同种异基因小鼠皮肤移植耐受诱导中的作用。方法 在“细胞后环磷酰胺”方法的基础上输注供体脾细胞或去除了T细胞的供体脾细胞 ,然后进行皮肤移植。观察皮肤存活情况并探讨耐受形成机制。结果 脾细胞输注后供体皮肤存活明显延长 ,而去除T细胞后皮肤存活不能被延长。克隆不应答、抑制细胞和嵌合体的形成等机制都参与了耐受形成。结论 供体脾脏中的T细胞在同种异基因小鼠皮肤移植耐受诱导中具有重要作用 ,它可能和宿主中克隆不应答的发生。 Objective To explore the role of T cells in donor spleen in the induction of murine skin allografts tolerance. Methods The donor spleen cells (SPC) or T cell depleted donor SPC were injected into the recipients which had been treated by'cells followed by CP system', after which skin grafting was performed. Then the survival time of skin grafts was observed and the tolerance mechanisms were investigated.Results The survival time of skin grafts in mice which received donor SPC infusion was significantly longer than that of control, while the survival time was not significantly prolonged in mice that received T cell depleted donor SPC infusion.Clone anergy, suppressive cells and chimerism were probably involved in the formation of tolerance.Conclusion Allogeneic T cells in donor spleen play an important role in the induction of skin allografts tolerance. They may correlate with the formation of clone anergy, the production or activation of suppressive cells and the maintainance of chimerism in the hosts.
