免疫缺陷鼠重建免疫后抑制移植瘤生长的研究

Research on the inhibition of the growth of grafted tumor on SCID mice after immune reconstitution

目的 研究严重联合免疫缺陷 (severitycombinedimmunodeficiency,SCID)鼠在转移正常鼠淋巴细胞后免疫重建的功效及其抑制移植瘤生长的机理。方法 将肝癌细胞HHCC 972 4接种到免疫重建SCID鼠 ,观察移植瘤生长情况、CD4CD8计数及其比值、免疫球蛋白定量、脾细胞毒试验以及癌细胞凝集试验。结果 ①重建免疫功能后SCID鼠对移植瘤有排斥作用。②SCID小鼠免疫重建后 4～ 6周CD4及CD8计数分别为 0 .1 2、0 .1 6 ,CD4 CD8为 0 .75。③免疫重建SCID小鼠产生较高水平的免疫球蛋白能对癌细胞产生凝集效应。④Balb c鼠和CBA N鼠的脾细胞对癌细胞排斥作用与免疫重建的SCID鼠相比较 ,后者仍显得较弱。结论 严重联合免疫缺陷SCID鼠可以通过免疫重建 。

Objective To investigate the growth inhibition of grafted tumor on SCID mice after immune reconstitution by infusion of the immune lymphocytes of the normal mice, and it's mechanism.Methods The hepatocarcinoma cells HHCC 9724 were inoculated into the immune reconstituted SCID mice . The indexes were surveyed on these mice as follows: growth condition of the grafted tumor, determination of the percentage of CD4 and CD8 and the ratio of CD4/CD8, immunoglobulin concentration of sera, cytotoxicity test of the spleen cells and serum agglutinations with tumor cells. Results ①Less grfted tumor was observed in the immune reconstitution SCID mice. ② The percentage of the CD4 and CD8 was 0.12 and 0.16 respectively and the ratio of the CD4/CD8 was 0.75 in the immune reconstitution of SCID mice 4～6 weeks later.③The immunoglobulin of the immune reconstitution SCID mice was increasing in high concentration.The immunoglobulin test showed the agglutination reaction with carcinoma cells could be observed.④The toxicity test of the mice spleen cells showed that the toxicity against the carcinoma cells in Balb/c mice and CBA/N mice was stronger than that found in immunoreconstituted SCID mice.Conclusion The immune function was reconstituted in immune deficiency SCID mice and the growth inhibition of the grafted tumor was observed.
