PPD对T_(H1)细胞的在体调节及对卵蛋白致敏小鼠气道炎症的影响 被引量:2

摘要 目的 探索结核菌素纯蛋白衍生物 (purifiedproteinderivative ,PPD)对卵蛋白 (ovalbumin ,OVA)致敏小鼠肺组织TH1 细胞的在体调节和对气道炎症的作用。方法 取 37只Balb c小鼠 ,雌雄兼有 ,6~ 8周。OVA雾化连续 1 0d ,每次雾化2 0min ,复制OVA致敏模型。观察记录小鼠耗氧量、肺组织淋巴细胞反应和支气管肺泡灌洗液 (bronchoalveolarlavagefluid,BALF)中炎性细胞变化。结果 OVA致敏组小鼠肺组织CD4+T淋巴细胞增加 ,CD8+T淋巴细胞无明显变化 ,主要表现为γ IFN阴性CD4+T淋巴细胞的增加 ,γ IFN+ CD4比例降低。PPD治疗后 ,肺组织CD4+淋巴细胞降低 ,但γ IFN+细胞明显增加。OVA致敏小鼠消耗氧增加 ,PPD治疗后降低。结论 使用本实验体系PPD可上调TH1 Objective To explore the in vivo regulation of PPD on T H1 cells and its effect on airway inflammation in ovalbumin sensitized mice.Methods Male and female Balb/c mice between 6 and 8 weeks of age were used in this study. Immunization was carried out utili zing a daily 20 min exposure to OVA (gradeⅢ,Sigma ) over a 10 days period by airway inhalation. T lymphocyte reaction, oxygen consumption and airway inflammation were investigated. Results There was a significant increase of γ IFN negative CD4 +T lymphocytes, oxygen consumption and eosinophils in OVA sensitized mice. PPD caused decrease of γ IFN negative CD4 +T lymphocytes and oxygen consumption ,but did not have any effect on eosinophils. Conclusion PPD could in vivo upregulate T H1 lymphocytes and decrease oxygen consumption in this animal model.
