摘要
目的 自鸽视盖克隆CDC10全长cDNA。 方法 建立鸽左侧单眼剥夺模型 ,采用cDNA末端快速扩增法和RNA转录物转换机制技术分别克隆鸽CDC10基因的 3′和 5′半分子并进行序列分析。 结果 鸽CDC10cDNA全长为 2 32 9bp ,开放阅读框为 12 5 7bp ,推测编码 4 19个氨基酸残基 ,氨基酸水平与人CDC10有 87%的同源性。
Objective To clone and sequence full\|length cDNA of CDC10 of pigeon. Methods The left monocular deprivation model of pigeons was established.Using techniques of RNA amplification of cDNA end(RACE) and switch mechanism at RNA transcript(SMART),3′half molecular and 5′half one were cloned respectively and sequenced subsequently. Results Full\|length cDNA of CDC10 of pigeon is 2?329?bp, its open reading frame is 1?257?bp,postulating encoding 419 amino acid residues and showing 87% homology with human CDC10 at amino acid level.Conclusion\ The cloned full\|length cDNA of CDC10 of pigeon may be helpful to further study its function during vision development\;[
