妊娠高血压综合征患者外周血及胎儿脐血端粒酶活性的初步研究 被引量:2

摘要 目的 :探讨外周血细胞端粒酶活性与妊娠高血压综合征 (简称妊高征 )的关系。 方法 :用端粒酶PCR ELISA方法分别测定 30例健康未妊娠妇女 (健康未妊娠组 )、4 8例正常单胎初产妇 (正常妊娠组 )及 74例妊高征患者 (妊高征组 )外周血及胎儿脐血端粒酶活性。 结果 :①妊高征组外周血单个核细胞的端粒酶活性阳性率为78.38% ,显著高于正常妊娠组 (14 5 8% )及健康未妊娠组 (6 .6 7% ) (P <0 .0 1) ,正常妊娠组与健康未妊娠组间无显著差异。②妊高征组脐血单个核细胞端粒酶活性 (5 8.90 % )较正常妊娠组 (10 .4 2 % )明显增高 (P <0 .0 5 )。③端粒酶活性与疾病严重程度呈正相关。④妊高征患者经解痉、降压治疗 5~ 8天后 ,与治疗前端粒酶活性差异无统计学意义。 结论 :端粒酶是细胞增生活性分子水平的标志物 ,外周血单个核细胞端粒酶活性增高可能与胎盘床子宫螺旋动脉血管平滑肌细胞增生有关 。 Objectives:To investigate the telomerase activity of peripheral blood mononuclear cells from patients with pregnancy induced hypertension(PIH). Methods:Maternal venous blood were collected from 30 non pregnant healthy women,48 normotensive pregnant women in the third trimester and 74 patients with PIH, and umbilical venous blood were collected from 48 newborns in normotensive pregnant group and from 73 newborns in the PIH group, respectively. The telomerase activity of peripheral and umbilical blood mononuclear cells was measured with telomerase PCR ELISA assay. Results:①The telomerase activity of maternal peripheral blood mononuclear cells in PIH group was significantly greater than that in normotensive pregnant group and non pregnant healthy group, while were no significant difference between the two control groups.②The telomerase activity of umbilical blood mononuclear cells in PIH was significantly greater than that in normotensive pregnant group.③The telomerase activity was positively correlated with the severity of hypertension.④There was no difference in telomerase activity before treatment and 5~8 days after treatment. Conclusions:Telomerase was a marker of cellular proliferation activity in molecular level, the elevated telomerase activity may be associated with the proliferation of vascular smooth muscle cells(VSMC) of placental bed spiral arteries. The result showed that elevated telomerase activity in patients with PIH might take part in the pathogenesis of PIH.
