摘要
目的 :探讨涎腺肿瘤与c myc,p5 3 ,bcl 2 ;PCNA基因及细胞凋亡的关系。 方法 :采用脱氧核糖核酸末端转移酶介导的dUTP缺口末端标记 (TUNEL)技术 ,免疫组织化学染色和HE染色对 18例多形性腺瘤和5 2例恶性涎腺肿瘤中c myc ,p5 3 ,bcl 2 ,PCNA基因及细胞凋亡进行观察 ,分析c myc,p5 3 ,bcl 2 ,PCNA基因及细胞凋亡与涎腺肿瘤的相关性。结果 :p5 3与bcl 2基因呈正相关 (P <0 0 5 ) ,c myc与 p5 3 ,bcl 2 ,PCNA基因呈正相关 (P <0 0 5 ) ,PCNA基因与细胞凋亡呈正相关 (P <0 0 5 ) ,而细胞凋亡与c myc ,p5 3 ,bcl 2基因间无相关性 (P >0 0 5 )。Fisher判别分析结果表明 ,用c myc ,p5 3 ,bcl 2 ,PCNA基因及细胞凋亡对涎腺肿瘤进行判别 ,判别符合率为 84 3 %。结论 :涎腺肿瘤是多种癌基因共同作用的结果 ,也进一步证实涎腺肿瘤中存在细胞增殖及细胞凋亡的紊乱 ;c myc,p5 3 ,bcl 2 。
Objective:To explore the relationship between c-myc,p53,bcl-2,PCNA and apoptosis with salivary gland tumor.Methods:Apoptosis was detected in histological sections by the terminal deoxynucleotidyl transferase-mediated dUTP nick and labelling (TUNEL) technique,c-myc,p53,bcl-2 and PCNA were detected by immunohistochemical staining in 18 cases of pleomorphic ademoma and 52 cases of salivary gland tumor.Results:p53 was correlated with bcl-2( P <0 05),c-myc was correlated with p53,bcl-2,PCNA( P <0 05),PCNA was correlated with apoptosis( P <0 05),but apoptosis was not related to c-myc,p53,bcl-2( P >0 05).Fisher discriminant analysis result showed 84 3% of original grouped cases correctly classified.Conclusion:Several gene cooperated in salivary gland tumors,it further indicated that disorder of apoptosis and cell proliferation play an important role in tumorigenesis of salivary gland tumors,examination of c-myc,p53,bcl-2,PCNA and apoptosis were found to be useful in distinguishing benign and malignant salivary gland tumor.
