川芎嗪对急性放射损伤小鼠骨髓血管内皮细胞生长因子表达水平的影响 被引量:8

Effects of tetramethylpyrazine(川芎嗪) on the expression of vascular endothelial growth factor in bone marrow in acute irradiation injured mice

摘要 目的 :探讨川芎嗪对急性放射损伤小鼠骨髓中血管内皮生长因子 (VEGF)表达水平的影响及其促进骨髓造血恢复的可能机制。方法 :5 6只清洁级昆明小鼠随机分为正常组、对照组和川芎嗪组。对照组和川芎嗪组一次性全身均匀照射γ射线后分别胃饲相同剂量的生理盐水和川芎嗪 ,分别于照射后第 3、7和 14日计数骨髓单个核细胞及骨髓组织中巨核细胞数 ,分析骨髓组织中造血组织面积及 VEGF表达水平。结果 :川芎嗪组于照射后第 3、7和 14日的骨髓单个核细胞计数、骨髓巨核细胞计数及骨髓造血组织面积均显著高于对照组(P<0 .0 5或 P<0 .0 1) ,川芎嗪组 VEGF的表达水平在第 3和 7日明显高于对照组 (P<0 .0 5或 P<0 .0 1) ,而第 14日恢复正常并明显低于对照组 (P<0 .0 5 )。结论 :川芎嗪可以促进急性放射损伤后造血功能的恢复 。 Objective:To evaluate the effect of tetramethylpyrazine(川芎嗪) on the expression of vascular endothelial growth factor(VEGF) in bone marrow tissue in acute irradiation injured mice and to explore the possible mechanism of tetramethylpyrazine on the recovery of hematopoiesis.Methods:The mice were randomly divided into three groups:normal group,irradiation injured group and tetramethylpyrazine group. After irradiation by 6 0 Gy 60 Co γray,each mouse was given orally by 2 mg tetramethylpyrazine twice a day for 13 days in tetramethylpyrazine group,and was given orally by equal quantity saline in irradiation injured group.On the 3rd,7th,14th day after irradiation,bone marrow mononuclear cells(BMNC)and megakaryocytes in bone marrow tissue were counted,the percentage of hematopoietic tissue and the expression levels of VEGF in bone marrow were evaluated.Results:On the 3rd,7th,14th day after irradiation,in tetramethylpyrazine group the BMNC,megakaryocytes counts and the percentage of hematopoietic tissue were higher than irradiation injured group ( P <0 05 or P <0 01).The VEGF expression levels in tetramethylpyrazine group were higher than irradiation group ( P <0 01 or P <0 05) on 3rd and 7th days,and lower than irradiation injured group on 14th day ( P <0 05).Conclusions:Tetramethylpyrazine can enhance VEGF expression in bone marrow in acute irradiation injured mice,which may be one of mechanisms of tetramethylpyrazine accelerating the recovery of hematopoiesis of mouse bone marrow.
