摘要
目的 :观察以线粒体膜电位 (MMP)标记法检测新风胶囊对佐剂性关节炎 (AA)大鼠滑膜及胸腺细胞凋亡的影响 ,探讨新风胶囊的作用机制。方法 :采用弗氏完全佐剂制成 AA大鼠模型 ,设立正常对照组、模型对照组、甲氨喋呤 (MTX)对照组、雷公藤多甙 (TPT)对照组和新风胶囊 (XFC)治疗组。记录各组大鼠的关节炎指数 ,用线粒体膜电位 (MMP)标记法测定各组大鼠滑膜及胸腺细胞凋亡率。结果 :与治疗前相比 ,XFC组、TPT组及 MTX组治疗后大鼠的关节炎指数均显著降低 (P<0 .0 5或 P<0 .0 1) ;模型对照组滑膜及胸腺细胞凋亡率显著低于正常对照组 (P<0 .0 5 ) ,XFC组细胞凋亡率显著高于模型对照组、TPT组及 MTX组 (P<0 .0 5或P<0 .0 1)。结论 :XFC与 MTX、TPT一样能降低 AA大鼠关节炎指数 ,在促进线粒体膜电位介导的滑膜及胸腺细胞凋亡方面 XFC优于 MTX和 TPT。促进滑膜细胞凋亡 ,抑制滑膜增生以及促进胸腺细胞凋亡 ,抑制自身免疫反应可能是 XFC降低关节炎指数。
Objective:To observe the effect of Xinfeng capsule(XFC,新风胶囊) on the apoptosis of the synoviocyte and thymuslymphocyte of adjuvant arthritis (AA) rat by the mitochondrial membrane potential (MMP).Methods:Model rats were established by the Freund's complete adjuvant.The rats were randomly divided into five groups:normal control,model control,methotrexate (MTX) control,triperygium wilfordii polycoride tablet (TPT) control,XFC treated.The arthritis index (AI) was recorded.The apoptotic percentage of the synoviocyte and thymuslymphocyte of AA were measured by flow cytometry (FCM) using MMP. Results: XFC,MTX and TPT could decrease the AI ( P <0 05 or P <0 01).Compared with the normal control, apoptotic percentage of the synoviocyte and thymuslymphocyte of model control was decreased ( P <0 05). Compared with the MTX,TPT and model control,apoptotic percentage of XFC was significant increased ( P <0 05 or P <0 01). Conclusions:XFC can decrease the AI like MTX,TPT.But it is superior to MTX,TPT in increasing the apoptosis of the synoviocyte and thymuslymphocyte of AA.It may be the mechanism for XFC to decrease AI that promote the apoptosis of synoviocyte and thymuslymphocyte, restrain the synovial membrane from hyperplasia and autoimmune reaction.
