摘要
目的 研究 1 甲基 4 苯基 1,2 ,3,6 四氢吡啶 (1 methyl 4 phenyl 1,2 ,3,6 tetrahydropyridine ,MPTP)诱导的黑质纹状体系统神经递质代谢相关基因表达的变化。方法 通过MPTP注射诱导C5 7BL小鼠神经元损伤 ,利用逆转录PCR方法检测酪氨酸羟化酶 (TH)、单胺氧化酶B(MAO B)、多巴胺转运体 (DAT)、胆碱乙酰基转移酶 (ChAT)以及谷氨酸脱羧酶 (GAD6 5 )五种神经递质代谢相关基因的表达变化。结果 在小鼠黑质与纹状体中 ,MPTP导致TH基因表达显著降低以及GAD6 5基因表达的上升。MAO B、DAT、ChAT基因的表达变化在黑质与纹状体则有所不同 ,在黑质MPTP导致MAO B与ChAT基因表达的上升以及DAT基因表达下降 ,在纹状体MPTP导致MAO B与ChAT基因表达下降 ,而对DAT基因表达没有明显影响。结论 MPTP对不同神经递质代谢相关基因表达有不同的影响 。
Objective To investigate the gene expression of neurotransmitter metabolic enzymes in substantia nigra and corpus striatum of 1?methyl 4?phenyl 1,2,3,6?tetrahydropyridine(MPTP) treated mice.Methods The nigrostriatal dopaminergic pathway of C57BL mice was damaged by administration of MPTP.The gene expressions of tyrosine hydroxylase(TH),monoamine oxidase(MAO) B dopamine transporter(DAT),choline acetyltransferase(ChAT)and Glutamate decarboxylase(GAD) in substantia nigra and corpus striatum of MPTP?treated C57BL mice were detected by reverse transcriptional ploymerase chain reaction(RT?PCR).Results The expression of TH gene was decreased in the substantia nigra and corpus striatum of MPTP?treated mice, the expressions of MAO?B and ChAT genes were increased in substantia nigra and slightly decreased in corpus striatum,the expressions of DAT gene was decreased in substantia nigra and showed no significant difference in corpus striatum.Conclusion The effect of MPTP on expression of TH,MAO?B,DAT,ChAT and GAD genes are different in MPTP?treated mice, and are specific in different brain regions.
