4例老年男性肺老化相关基因谱的研究

摘要 为了解老化过程中人类肺部基因表达谱改变 ,作者从老年和成年人正常肺组织提取总RNA ,用基因芯片技术比较基因表达差异。结果显示 ,研究的 8192条基因中有 87(1 0 6 % )条表达差异在 1 6倍以上 ,4 7(0 5 7% )条上调 ,4 0 (0 4 9% )条下调。上调为主的有 :细胞骨架和胞外基质、炎性应激反应、肿瘤相关、信号转导等 ;下调为主的有 :DNA/RNA合成修复、蛋白质合成修饰、能量代谢等 ;细胞周期与增殖相关基因上下调比例相当。提示老年人肺部基因表达谱发生了明显改变 ,与肺老化改变特征相符。 To understand changes in gene expression levels occurred in the process of aging in human lung, cDNA microarray technology was used to compare gene expression profile in lung of the elderly with that of the adult. Comparing 8 192 genes expression levels in lung of aged men with those in adults, 87 (1 06%) genes changed at least 1 6 fold. The expression of 47 (0 57%) genes was up regulated and that of 40 (0 49%) genes was down regulated. Those genes with increased expression level in aging lung mainly related to cytoskeleton or extracellular matrix, inflammation or stress response, tumor, signal transduction and so on. Those genes with decreased expression levels mainly related to DNA/RNA synthesis or repair, protein synthesis or processing, and energy metabolism. The changes of gene expression related to cell cycle or proliferation played an inhibitory role in them. It is suggested that gene expression in aging lung changes obviously, and the changes fit the character of aging lung.
