摘要 介绍了一种大规模提取高质量果蝇基因组DNA的方法,该法不需液氮研磨,不需匀浆转移,不需高速离心,全部操作在3个1.5μL离心管中完成,所以特别适用于对不同果蝇品系等多样本的大规模提取.以本法提取的DNA为模板建立了RAPD-PCR分析的最佳反应体系,获得了稳定而理想的扩增效果,特别适用于小型昆虫的群体遗传多样性研究. A method large-scale extraction of high quality genomic DNA from Drosophila is reported. This method need not grinding flies with liquid nitrogen, transferring the serosity and high speed centrifuging, all the operations are finished in three 1.5?μL micro-tubes .which is most applicable for large-scale DNA extraction from different samples. Using the DNA extracted by this method as template, established the optimum conditions for RAPD-PCR(random amplified polymophic DNA-polymorase chain reaction), which can be applied to genetic diversity studies of little insects.
