摘要
在已达到要求的温育时间范围内 ,选择最佳温育时间 ,稳定剂量反应曲线 ,使结果稳定、可靠。方法为 :(1)随机取 30份肝病患者血清 ,在 4℃分别温育 18h、2 0h、2 3h ,血清测定层粘连蛋白 (laminin ,LN)浓度 ,并对三个不同的温育时间所得的值进行显著性检验 ;(2 )对温育 18h、2 0h、2 2h、2 4h分别为四批的标准曲线所得的质控指标进行分析 ;(3)对不同温育时间的回收率及浓度进行分析 ;(4)随机取 2 0份肝病患者血清 ,每三批各做 2 0份 ,于温育 2 0h ,对其所得的值进行显著性检验 ,以观察批间误差。结果表明 :(1)三个不同温育时间的 30份血清LN测定值的t检验有显著性差异。P值分别小于 0 .0 1和小于 0 .0 5 ;(2 )不同温育时间的标准曲线B0 /T(% )、NSB/T(% )、相关系数 (r)的均值良好 ,无明显差异 ;温育 2 0h的ED50 的均值为 118.4ng/mL(范围 113.9- 12 3.8ng/mL) ,在正常参考范围内 (药盒正常参考值为 115 .7± 17.3ng/mL)。其它三个温育时间的ED50 均超也正常参考范围 ;(3)温育低于及高于 2 0h回收率均低于温育 2 0h。温育 2 0h的回收率最好 ,为 95 .2 %。 (4)随机取 2 0份肝病患者血清 ,三批温育 2 0h的批间结果均无显著性差异 ,P值均大于 0 .0 5。实验结果表明 ,本实验室测定LN在 4℃时 。
To select the optimal incubation time in the extent of required time, to stabilize the dose-response curve, our results were stable and reliable. Methods were:(1)We randomly chose 30 serum of hepatitc patients, which were incubated at 4℃ after 18h, 20h and 23h respectively, then serum levels of laminin were measured and the statistical significance was tested;(2)Quality control indexes of standard curves of four batches each after 18h?20h?22h and 24h incubation were analyzed;(3)The recovery rate and density of different incubation periods were analyzed;(4)20 serum of hepatic patients was selected and was divided into three batches incubated for 20h the statistical significance was tested to observe the errors between batches. Results were: (1)There were significant differences among the laminin levels of 30 samples incubated for three different periods( P <0.01, P <0.05);(2)Standard curves, B 0/T?NSB/T?coefficient of correlation were all good; The average of ED 50 in incubation for 20h was 118.4ng/mL, within normal reference range (The normal reference value of kit is 115.7±17.3ng/mL).ED 50 in the other three incubation periods all exceeded the normal reference range;(3)The recovery rates of incubation above or below 20℃ were all lower than that of incubation at 20℃. The latter was best(95.2%);(4)There were no significant difference between three bathes incubated for 20h( P >0.05).Results showed that the optimal incubation time was 20h for measured laminin at 4℃ in our laboratory. It suggested that people explore the optimal incubation periods according to their own conditions.
