干扰素-α对THP-1细胞TLR7表达的影响

摘要 目的探讨干扰素-α(Interferon-α,IFN-α)对人急性单核细胞白血病细胞系(THP-1)Toll样受体7(TLR7)表达的影响。方法以IFN-α、LPS处理THP-1细胞,MTT法检测细胞增殖情况;苏木精-伊红(HE)染色观察细胞形态学;反转录-PCR、Western blot方法检测细胞TLR7 mRNA的表达和蛋白表达。结果与对照组相比,IFN-α对细胞增殖具有明显抑制作用(P<0.05),IC50为2000 U/L;IFN-α作用THP-1细胞24 h、48 h后,细胞生长受到不同程度的抑制,可见细胞部分浮起、细胞变圆、体积缩小、核深染等改变;IFN-α组TLR7 mRNA和蛋白表达量明显上升(P<0.05)。结论 IFN-α抑制THP-1细胞生长,可能与上调TLR7基因及蛋白表达有关。 Objective To investigate the effect of interferon-alpha(IFN-α) on the expression of Toll-like receptor 7(TLR7) in human acute monocytic leukemia cell line(THP-1). Methods THP-1 cells were treated with IFN-α and LPS, and the proliferation of THP-1 cells was detected by MTT. ResultsCompared with the control group, IFN-alpha significantly inhibited the proliferation of THP-1 cells(P<0.05), IC50 was 2000 U/L. After IFN-alpha treated THP-1 cells for 24 and 48 hours, the growth of THP-1 cells was inhibited. Some changes were observed such as cell buoyancy, cell rounding, cell shrinkage and nuclear hyperchromatism et al. And the expression of TLR7 gene and protein in IFN-alpha group increased significantly(P<0.05). Conclusion IFN-alpha inhibits the growth of THP-1 cells, which may be related to the up-regulation of TLR7 gene and protein expression.
