c-Jun氨基末端激酶选择性抑制剂对哮喘小鼠肺组织IL-6表达的影响

The effect of selective JNK inhibitor on IL-6 in the lung of the asthmatic mice

目的探讨c-Jun氨基末端激酶选择性抑制剂(SP600125)对哮喘小鼠肺组织白介素-6(IL-6)表达的影响。方法 30只BALB/c小鼠随机分为对照组、哮喘组和SP600125组,制作哮喘模型及干预处理后,处死小鼠,取肺组织,采用免疫组织化学方法和western blot方法检测各组小鼠肺组织内IL-6的表达。结果免疫组化结果显示,哮喘组小鼠肺组织IL-6表达的平均光密度值为(0.71±0.11),显著高于对照组(0.23±0.03),P<0.01;而SP600125组小鼠肺组织IL-6表达的平均光密度值为(0.46±0.07),与哮喘组相比明显降低,P<0.01。western blot结果显示,哮喘组小鼠肺组织IL-6表达的平均光密度值为(0.62±0.09),显著高于对照组(0.16±0.02),P<0.01;而SP600125组小鼠肺组织IL-6表达的平均光密度值为(0.37±0.05),与哮喘组相比明显降低,P<0.01。结论 c-Jun氨基末端激酶选择性抑制剂能降低哮喘小鼠肺组织IL-6的表达。

Objective To study the effect of selective c-Jun N-terminal kinase(JNK) inhibitor on interleukin-2(IL-6) in the lung of the asthmatic mice. Methods 30 BALB / c mice were randomly divided into control group, asthmatic group and SP600125(selective JNK inhibitor)group on average. The asthmatic mice model was established by the inhalation of ovalbumin,and all animals were sacrificed after treatment. The expression of IL-6 in lung of mice was observed by immunohistochemistry and western blot methods. Results The mean optic density(MOD) of IL-6 was significantly higher in lung of the asthmatic mice than in thelcontrol group(0.71±0.11 vs 0.23±0.03, P < 0.01) by means of immunohistochemistry; compared with the asthmatic group, The MOD of IL-6 was significantly lower(0.46±0.07 vs 0.71±0.11, P < 0.01). The mean optic density(MOD) of IL-6was significantly higher in lung of the asthmatic mice than in thelcontrol group(0.62±0.09 vs 0.16±0.02, P < 0.01) by means of western blot method; compared with the asthmatic group, the MOD of IL-6 was significantly lower(0.37±0.05 vs 0.62±0.09, P <0.01). Conclusion Selective JNK inhibitor could down-regulate the expression of IL-6 in the lung of the asthmatic mice.
