行为学训练对缺血性脑损伤大鼠脑组织TNF-α和TNF-αmRNA表达的影响

摘要 目的研究大鼠脑缺血再灌注后对肿瘤坏死因子-α(TNF-α)和TNF-αmRNA表达的影响,探讨穿梭箱行为学训练对脑缺血再灌注大鼠脑组织TNF-α和TNF-αmRNA的调节作用及机制。方法将54只Wistar大鼠随机分为训练组;缺血再灌注组及正常对照组,每组18只。应用线栓法制作大鼠局灶性脑缺血再灌注模型,训练组于造模ld后开始给予穿梭箱训练,采用免疫组化和RT-PCR法检测大脑皮质内TNF-α和TNF-αmRNA的表达。结果正常对照组大鼠大脑皮质内TNF-α和TNF-αmRNA少量的表达,缺血再灌注后高于正常对照组,训练组大脑皮质内TNF-α和TNF-αmRNA阳性表达较缺血再灌注组明显减少。结论穿梭箱训练下调脑缺血诱导的TNF-α和TNF-αmRNA表达,对脑缺血再灌注大鼠皮质神经元的具有保护作用。 Objective To investigate the expression of tumor necrosis factor-alpha(TNF-α) and TNF-αmRNA in cortex after cerebral ischemia and reperfusion in rats, explore the regulative effects and mechanism of Behavior Training to TNF-α and TNF-αmRNA on ischemia brain tissue. Methods 54 wistar rats were randomized into training groups、 ischemia and reperfusion groups and control group. Behavior training were performed at lst day after the ischemia and reperfusion. The model of middle cerebral arteries occlusion(MCAO) were performed with intraluminal filament blockade. the expression of TNF-α and TNF-αmRNA in cortex was detected with immunohistochemical and RT-PCR method. Results The expression of TNF-αand TNF-αmRNA is certain in the normal cortex of rats. The expression of TNF-α and TNF-αmRNA increased after model group. However, the expression of TNF-α and TNF-αmRNA had obviously decreased in training groups compared with that in ischemia and reperfusion groups. Conclusion The results indicate that training groups participate in the regulation of expression of TNF-α and TNF-αmRNA in ischemic neurons.
