摘要
目的分析硫化氢H2S合成酶基因表达水平与人神经胶质瘤分化与生存率的相关性。方法从肿瘤基因组图谱(TCGA)的脑低分化神经胶质瘤(BLGG)模块收集神经胶质瘤患者共530例,其中男患者291例,女患者238例,1例性别不详。患者年龄范围14~87岁,确诊为脑低分化神经胶质瘤Ⅱ级248例,Ⅲ级265例。结果在神经胶质瘤Ⅱ级患者,脑H2S合成酶胱硫醚β合成酶(CBS)的基因表达水平明显高于Ⅲ级患者(P<0.05)。CBS基因表达水平高于或等于11.63的神经胶质瘤患者的生存率显著高于CBS基因表达水平低于11.63的患者。CBS基因表达水平与神经胶质瘤患者的年龄呈负相关。在神经胶质瘤患者中,另一个H_2S合成酶3硫基丙酸硫基转移酶(MPST)的基因表达水平与CBS的基因表达水平呈正相关。MPST基因表达水平大于10.27的神经胶质瘤患者的生存率明显高于低表达(<9.51)组的患者(P<0.05)。结论 H2S合成酶的基因表达水平与人神经胶质瘤的分化及生存率密切相关。
Objective To analyze the relativity of level of hydrogen sulfide(H2 S)synthase gene expression to tumor grade and survival probability in patients with glioma. Methods The data of 530 cases of glioma(including male291,female 236,NA 1)were collected from the brain lower grade glioma(BLGG)station of The Cancer Genome Atlas(TCGA). The patient age was from 14 years old to 87 years old. Among the patients introduced in this study,248 cases were diagnosed grade Ⅱ tumor,265 cases grade Ⅲ tumor. Results The expression level of cystathionine β-synthase(CBS)gene was higher in the glioma patients with grade Ⅱ tumor than in the ones with grade Ⅲ tumor(P<0.05). The survival probability of the patients having high expression level of CBS gene(≥ 11.63)was significantly higher than the one of the patients having lower expression of CBS gene(<11.63). The expression level of CBS gene was negative relativity to age of the patients. There was a positive relativity between expression level of another H2 S synthase(3-mercaptopyruvate sulfurtransferase;MPST or 3-MST)gene and the one of CBS gene. The survival probability of the patients having MPST gene expression level higher than 10.27 was higher than the one of patients having lower expression level(<9.51)(P<0.05).Conclusion The expression level of H2 S synthase gene is markedly relative to tumor grade and survival probability in the patients with glioma.
