摘要
目的观察体外状态格拉默(GA)免疫后刺激机体产生的细胞因子对神经干细胞增殖及分化的影响。方法分离和培养新生SD大鼠脑室下区神经干细胞。4周龄雄性SD大鼠10只随机分为格拉默组(GA免疫组)及对照组,每组5只。格拉默免疫结束后2周,GA免疫组及对照组SD大鼠静脉血清分别加至已培养的脑室下区神经干细胞,培养3~4 d后计数增殖的神经球数目及直径大小。细胞分化完成后进行免疫细胞荧光染色。结果加入格拉默刺激后的血清神经干细胞球数目较对照组增加40%,且神经球直径较对照组大。神经球经免疫细胞化学鉴定为神经干细胞。格拉默免疫组血清较对照组血清更加促进神经干细胞向神经元方向分化。结论格拉默免疫通过特异性T细胞激活分泌的细胞因子有助于神经干细胞的增殖及向神经元方向的分化。
Objective To detect whether Glatiramer acetate(GA)may reverse cognitive deficits from cranial-irradiated rat and to obverse the influence on neurogenesis. Methods Brains from postnatal SD rats(1-day old)were dissected to remove the olfactory bulb,cerebellum and brainstem. Rat blood was collected from control and GA animals into EDTAcoated tubes via intracardial bleed at time of two weeks after GA administration. And then,we observed that the number and size of neurospheres,and its differentiation formed from primary NPCs exposed to the above. Results The number of neurospheres formed after exposure of primary neural stem/progenitor cells(NPCs)cocultured with GA immuned serum We assayed,and observed a 40% increase in GA group when compared to vehicle. We then observed that the size of neurospheres in GA group significantly increased. Neurospheres were approved to be neural stem/progenitor cells(NPCs)by immunocytochemistry. GA immuned serum greatly promoted NPCs differentiate to neurons with respect to the vehicle control serum. Conclusion GA immunity may contribute to promote NPCs proliferate and differentiate to neurons with cytokines secreted by specific T-cells.
