RUNX2基因扩增与骨肉瘤组织上皮-间质转换和患者预后的关系

Relationship between RUNX2 gene and epithelial-interstitial transformation and prognosis in osteosarcoma patients

目的探讨RUNX2基因扩增与骨肉瘤组织上皮-间质转换和患者预后的关系,旨在为分析RUNX2在介导骨肉瘤恶性生物学行为进展中的作用。方法采用回顾性研究方法,选取我院诊治的骨肉瘤患者60例,同期选取癌旁组织30例,采用免疫组化Envision法检测RUNX2基因的蛋白水平,同时检测上皮-间质转换标记物E-cadherin和Vimentin的表达情况,采用相关分析RUNX2与上皮-间质转换的关系,同时采用生存曲线分析RUNX2基因表达与患者中位生存时间的关系。结果 RUNX2在骨肉瘤组织中的表达阳性率为68.33%,明显高于癌旁组织,差异有统计学意义(P<0.05);在41例RUNX2阳性组织中,上皮-间质转换发生率明显升高,表现为E-cadherin表达降低和Vimentin表达增加,明显高于RUNX2阴性组织(P<0.05)。RUNX2阳性组中患者的中位生存时间为(27.33±7.12)月,3年生存率为19.51%(8/41),明显低于RUNX2阴性组,差异有统计学意义(P<0.05)。结论骨肉瘤组织中存在RUNX2基因异常扩增表达,并参与介导了上皮-间质转换的发生,后者与化疗耐药和患者预后关系密切。

Objective To study the relationship between RUNX2 gene and epithelial-interstitial transformation and prognosis in osteosarcoma patients,aims at analyzing RUNX2 in mediating the osteosarcoma malignant biological behavior progress. Methods Selected from 60 patients with osteosarcoma and 30 cases tissue adjacent to carcinoma,using the immunohistochemical envision method to detect the RUNX2 gene protein levels,and detecting epithelial mesenchymal transition markers E-cadherin and Vimentin expression. Correlation between RUNX2 and epithelial-qualitative transformation were analyzed. at the same time,the survival curves analysis about RUNX2 gene expression and the median survival time of patients were also uesd. Results The positive rate of expression of RUNX2 in osteosarcoma tissue was 68.33%,which was significantly higher than that of adjacent tissues(P<0.05). In 41 cases of RUNX2 positive tissues,the incidence of epithelialmesenchymal transformation was significantly increased,showing decreased expression of e-cadherin and increased expression of Vimentin,and it significantly higher than that of RUNX2 negative tissues(P<0.05). The median survival time of patients in the RUNX2 positive group was(27.33+7.12)months,and the three-year survival rate was 19.51%(8/41),significantly lower than that of the RUNX2 negative group(P<0.05). Conclusion The abnormal expression of RUNX2 gene in osteosarcoma is involved in epithelial-mesenchymal transformation,which is closely related to chemotherapy resistance and prognosis of patients.
