卵巢癌组织中Oct-4与Sox-2的表达及意义 被引量:2

摘要 目的探讨转录因子Oct-4与Sox-2在卵巢癌组织中的表达及临床意义。方法选取36例卵巢癌组织和19例正常卵巢上皮组织标本,采取免疫组织化学方法和Western blot方法检测两组组织标本中Oct-4和Sox-2的蛋白表达水平,采用荧光定量PCR法检测两组组织标本中Oct-4、Sox-2 mRNA的表达量,分析卵巢癌组织Oct-4、Sox-2的蛋白表达与卵巢癌临床病理参数的关系。结果卵巢癌组织中Oct-4与Sox-2蛋白表达的平均光密度值显著高于正常卵巢上皮组织(P<0.01);卵巢癌组织中Oct-4与Sox-2 m RNA的相对表达水平亦显著高于正常卵巢上皮组织(P<0.01)。Oct-4与Sox-2在卵巢癌组织中的表达与卵巢癌的肿瘤分级密切相关(P<0.01),与患者年龄、病理类型和淋巴结转移无关(P>0.05)。结论卵巢癌组织中Oct-4和Sox-2的表达上调,并与卵巢癌的肿瘤分级密切相关。 Objective To investigate the expression and clinical significance of transcription factors Oct-4 and Sox-2 in ovarian cancer. Methods 36 cases of ovarian cancer tissue and 19 normal ovarian epithelial tissue specimens were selected. The protein expression of Oct-4 and Sox-2 of tissue specimens in two groups were detected by immunohistochemical method and Western blot method. The expression level of Oct-4 and Sox-2 mRNA of tissue specimens in two groups was detected by real time PCR. The relationship between the expression of Oct-4 and Sox-2 with the clinicopathological factors of ovarian cancer was analyzed. Results The mean optical density of Oct-4 and Sox-2 protein expression in ovarian cancer tissues was significantly higher than that of normal ovarian epithelial tissues(P<0.01),and the relative expression level of Oct-4 and Sox-2 mRNA in ovarian cancer tissues was also significantly higher than that of normal ovarian epithelial tissues(P<0.01). The expression of Oct-4 and Sox-2 in ovarian cancer tissues is closely related to the classification of ovarian cancer(P<0.01),and not related to age,pathological type and lymph node metastasis(P>0.05). Conclusion The expression of Oct-4 and Sox-2 is up-regulated in ovarian cancer tissues and is closely related to the tumor grade of ovarian cancer.
