摘要
目的研究槲皮素治疗糖尿病肥胖大鼠(Zucker diabetes fatty,ZDF)糖脂代谢紊乱及胆固醇的作用机制。方法成年雄性Wistar大鼠(90只)给予高糖高脂喂养,并辅以链脲佐菌素腹腔内注射建立糖尿病肥胖模型,随机分为对照组、模型组和实验组。实验组大鼠给予槲皮素治疗。模型构建8周后处死大鼠,测定大鼠空腹血糖(FBG),检测大鼠空腹胰岛素(FINS)、糖化血红蛋白(HbA1c),计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(HOMA-IR);测定大鼠胆固醇(TC)、游离脂肪酸(FFA)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C);HE染色观察测定大鼠胰腺组织学改变。结果与模型组相比,实验组大鼠TC、FFA、TG、LDL-C含量明显降低,HDL-C含量显著增高(P<0.05);实验组大鼠FBG、FINS、HbA1c、ISI和HOMA-IR含量明显降低(P<0.05);实验组大鼠血清内尿素氮(BUN)、肌酐(Scr)及24 h尿蛋白定量含量明显降低(P<0.05);实验组大鼠谷丙转氨酶(ALT)含量明显降低,过氧化氢酶(CAT),超氧化物歧化酶(SOD)和丙二醛(MDA)和谷胱甘肽过氧化物酶(GSHPx)含量明显升高(P<0.05)。结论槲皮素能够有降低糖尿病肥胖大鼠血糖和胆固醇、调节血脂紊乱,对糖尿病肥胖大鼠具有一定的治疗作用。
Objective To study the mechanism of quercetin on the metabolism of glucose and lipids and cholesterol in diabetic obese rats(ZDF). Methods Ninety adult male Wistar rats were fed high-sugar and high-fat diets and supplemented with intraperitoneal injection of streptozotocin to establish diabetic obesity models. They were randomly divided into three groups:control group,model group and experimental group. Rats in the experimental group were treated with quercetin. Rats were sacrificed 8 weeks after the model was constructed.Fasting blood glucose(FBG)was measured in rats,fasting insulin(FINS),glycosylated hemoglobin(HbA1 c),insulin sensitivity index(ISI)and insulin resistance index(HOMA-IR)were measured. TC,FFA,TG,HDL-C and LDL-C in rats;detection of rat pancreatic histology by HE method change.Results Compared with the model group,the cholesterol,TC,HDL,and HDL-C levels in the experimental group were significantly lower than those in the experimental group. High-density lipoprotein(LDL-C)Significant increase in content(P<0.05). Rats in the experimental group had fasting blood glucose(FBG),fasting insulin(FINS),HbA1 c,calculated insulin sensitive index(ISI),and insulin resistance index(HOMA-IR)significantly lower(P<0.05);The serum urea nitrogen(BUN),creatinine(Scr),and 24-hour urinary protein levels in the experimental group were significantly lower(P<0.05);the alanine aminotransferase(ALT)content in the experimental group was significantly reduced,catalase(CAT),SOD,MDA and GSH-Px levels were significantly increased(P<0.05). Conclusion Quercetin can reduce blood glucose and cholesterol,regulate blood lipid disorders in diabetic obese rats,and has a certain therapeutic effect on diabetic obese rats.
