核因子-κB p65在神经病理性疼痛大鼠脊髓中的表达定位 被引量:5

摘要 目的研究核因子-κB(NF-κB)p65在神经病理性疼痛大鼠脊髓背角中的表达与定位。方法建立大鼠坐骨神经慢性压迫损伤(CCI)模型;应用Western blot法检测CCI大鼠脊髓NF-κB p65的表达情况;采用免疫荧光染色双标法检测NF-κB p65蛋白在CCI大鼠脊髓背角内的亚细胞(神经元、星形胶质细胞及小胶质细胞)定位。结果 CCI大鼠的机械阈值较正常大鼠显著降低;神经损伤后,NF-κB p65表达量在脊髓组织中显著增高;NF-κB p65主要定位于脊髓背角神经元和星形胶质细胞,而在小胶质细胞中没有表达。结论 NF-κB p65在CCI大鼠脊髓中相对高表达并主要定位于脊髓背胶神经元和星形胶质细胞。 Objective To investigate the expression and localization of NF?κB p65 in spinal cord of rats with neuro?pathic pain.Methods Chronic constriction injury of the sciatic nerve was used to build neuropathic pain model.The expres?sion level of NF?κB p65 in spinal cord of CCI rats were detected by Western blot.The subcellular localization of NF?κB p65 in spinal dorsal horn of CCI rats was detected by immunofluorescence staining.Results The mechanical withdrawal thresh?old of CCI rats was significantly lower than that of normal rats.The expression level of NF?κB p65 was significantly increased after peripheral nerve injury.NF?κB p65 was demonstrated to localize in neurons and astrocytes.In contrast,NF?κB p65 was not expressed in microglia.Conclusion NF?κB p65 protein was highly expressed in spinal cord of CCI rats,and was localized principally in neurons and astrocytes.
