摘要
目的探讨下调YAP1蛋白对子宫内膜癌细胞增殖的影响和其相关的机制。方法运用慢病毒载体建立YAP1缺失的细胞株(HEC-1-B-shYAP)和阴性对照(HEC-1-B-NC),分别采用Real-time PCR及Western blot法检测细胞YAP1的表达。CKK-8方法检测细胞增殖能力的改变,Western blot法检测Wnt信号通路相关蛋白β-catenin的表达。结果成功构建YAP1稳定缺失的卵巢癌细胞株(HEC-1-B-shYAP1),与阴性对照组和空白对照组细胞相比,HEC-1-B-shYAP组细胞增殖能力明显减弱(P<0.01),同时β-catenin的表达明显降低(P<0.01)。结论 YAP1可促进子宫内膜癌的增殖,其机制可能Wnt-β-catenin信号通路有关。
Objective To investigate the effect of down?regulated YAP1 protein on the proliferation of endometrial cancer cells and its related mechanisms.Methods Cell lines(HEC?1?B?shYAP)and negative controls(HEC?1?B?NC)were established with lentivirus vector,Real?time PCR and Western blot were used to detect the expression of YAP1.CKK?8 was used to detect changes in cell proliferation capacity,while Western blot method was applied to detect the expression of Wnt signaling pathway related proteinsβ?catenin.Results The ovarian cancer cell line(HEC?1?B?shYAP1)with stable YAP1 deletion was successfully constructed.Compared with cells in the negative control group and the blank control group,the proliferation capacity of cells in the HEC?1?B?shYAP1 group was significantly decreased.At the same time the expression ofβ?catenin significantly decreased(P<0.01).Conclusion YAP1 can promote the proliferation of endometrial carcinoma,and its mechanism may be related to the wnt?yoky?catenin signaling pathway.
