LncRNA SNHG1通过吸附miR-199a-3p促进肾透明细胞癌细胞迁移和侵袭

Lnc RNA SNHG1 promotes the migration and invasion of renal clear cell carcinoma cells by adsorbing mir-199a-3p

目的探讨LncRNA SNHG1对肾透明细胞癌细胞侵袭及转移能力的影响,并探索潜在机制。方法利用划痕实验和Transwell实验检测SNHG1对肾透明细胞癌细胞迁移和侵袭能力的影响,利用双荧光素酶报告基因实验检测SNHG1是否可结合miR-199a-3p,利用实时荧光定量PCR检测SNHG1与miR-199a-3p表达的关系。结果抑制SNHG1的表达后,细胞迁移和侵袭能力均升高(P<0.01)。双荧光素酶报告基因实验结果显示SNHG1能够吸附miR-199a-3p(P<0.01)。实时荧光定量PCR实验结果显示抑制SNHG1的表达可促进miR-199a-3p的表达(P<0.01)。结论 SNHG1可通过吸附miR-199a-3p促进肾透明细胞癌细胞迁移和侵袭。

Objective To investigate the effects of LncRNA SNHG1 on the invasion and metastasis of renal clear cell carcinoma cells and explore the underlying mechanisms.Methods Wound healing assays and Transwell assay were used to detect the effect of SNHG1 on the migration and invasion of renal clear cell carcinoma cells.The dual luciferase reporter gene assay was used to detect whether SNHG1 can bind to miR-199a-3p.Real-time PCR were performed to detect the relationship between SNHG1 and miR-199a-3p.Results After inhibiting the expression of SNHG1,cell migration and invasion ability were increased.Dual luciferase reporter gene assay showed that SNHG1 could directly bind with miR-199a-3p.Real-time PCR results revealed that knockdown SNHG1 could promote the expression of miR-199a-3p.Conclusion SNHG1 could promote migration and invasion of renal clear cell carcinoma cells by sponging miR-199a-3p.
