摘要
目的探讨GAS5基因启动子甲基化在老年胃癌组织中的水平及其与临床病理特征的相关性。方法纳入2014年9月-2016年9月于我院诊断的100例胃癌患者。采用甲基化特异性PCR检测老年胃癌组织和癌旁组织中GAS5基因的甲基化水平。分析GAS5基因启动子甲基化水平与患者临床病理特征及预后的关系。结果老年胃癌组织中GAS5基因启动子存在高甲基化(P<0.05),但是癌旁组织中GAS5基因启动子的甲基化水平较低(P<0.05);GAS5基因启动子高甲基化与老年胃癌肿瘤的最大径和临床分期有相关性(P<0.05),而与性别和家族史无相关性(P>0.05);Cox回归分析发现肿瘤最大径≥5 cm、临床分期Ⅲ期及GAS5基因启动子高甲基化水平与老年胃癌患者生存不良预后相关(P<0.05)。结论 GAS5基因启动子在老年胃癌组织中高甲基化,且其与老年胃癌患者生存不良预后相关,是一种分析老年胃癌患者预后的潜在指标。
Objective To investigate the level of GAS5 gene promoter methylation in elderly gastric cancer and its correlation with clinicopathological features.Methods 100 patients with gastric cancer diagnosed in our hospital from September 2014 to September 2016 were included.Methylation-specific PCR was used to detect the methylation level of GAS5 gene in elderly gastric cancer tissues and adjacent tissues.The relationship between the methylation level of GAS5 gene promoter and clinicopathological features and prognosis was analyzed.Results GAS5 gene promoter was hypermethylated in elderly gastric cancer tissues(P<0.05),but the methylation level of GAS5 gene promoter was lower in adjacent tissues(P<0.05);GAS5 gene promoter hypermethylation and elderly gastric cancer There was a correlation between the maximum diameter of the tumor and the clinical stage(P<0.05),but no correlation with gender and family history(P>0.05).Cox regression analysis found that the tumor had a maximum diameter of≥5 cm,clinical stage III and GAS5 gene.Promoter hypermethylation level was associated with poor prognosis in elderly patients with gastric cancer(P<0.05).Conclusion GAS5 gene promoter is hypermethylated in elderly gastric cancer tissues,and it is associated with prognosis of elderly patients with gastric cancer.It is a potential index for analyzing the prognosis of elderly patients with gastric cancer.
