HPLC法测定化香虫茶中没食子酸和鞣花酸的含量 被引量:6

Determination of gallic acid and ellagic acid in Platycarya strobilacea insect tea by HPLC

摘要 目的建立双波长RP-HPLC法测定化香虫茶中没食子酸、鞣花酸质量分数。方法采用COSMOSIL C18-AR-Ⅱ色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%(体积分数) H3PO4,梯度洗脱,流速为1.0 mL/min,检测波长为269 nm(没食子酸)、254 nm(鞣花酸),柱温为25℃。结果没食子酸和鞣花酸分别在0.052~0.26、0.21~1.24μg范围内呈良好的线性关系,醇提物的没食子酸和鞣花酸平均回收率分别为98.03%、102.99%,RSD分别为1.10%、1.09%。水提物的没食子酸和鞣花酸平均回收率分别为98.85%、99.08%,RSD分别为2.74%、2.78%。结论本方法操作简便、准确、重复性良好,可用于化香虫茶中没食子酸、鞣花酸质量分数的测定。 Objective To determine the gallic acid and ellagic acid in the Platycarya strobilacea insect tea(PSI)by HPLC.Methods The separation was performed on the COSMOSIL C18-AR-Ⅱ(250 mm×4.6 mm,5μm)column,and acetonitrile-0.1%H3PO4 in gradient as mobile phase,the flow rate was 1.0 m L/min,and the detection wavelengths were set at 269(gallic acid),254 nm(ellagic acid),and the column temperature was 25℃.Results The linear ranges of gallic acid and ellagic acid were within 0.052-0.26,0.21-1.24μg respectively.The average recoveries for gallic and ellagic acid in ethanol extract of PSI were98.03%and 102.99%,RSD were 1.10%and 1.09%(n=5).The average recoveries for gallic and ellagic acid in water extract of PSI were 98.85%and 99.08%,RSD were 2.74%and 2.78%(n=5).Conclusion The method is simple,accurate,reproducible and suitable for the determination of gallic acid and ellagic acid in the Platycarya strobilacea insect tea.
