摘要
目的观察短链酰基辅酶A脱氢酶(Short-chain acyl-CoA dehydrogenase,SCAD)在老年自发性高血压大鼠慢性心力衰竭中的变化,探讨SCAD在老年自发性高血压大鼠慢性心力衰竭中的作用。方法选取20月龄雄性SHR及年龄匹配的WKY对照大鼠,分为WKY对照组和SHR实验组。检测大鼠超声心动图、血压、左室质量指数、心肌纤维化、细胞凋亡、SCAD mRNA和蛋白表达、心肌组织ATP含量、线粒体膜电位和肿胀程度。结果与WKY对照组比较,SHR组心脏的射血分数和缩短分数明显减少,心输出量显著降低,心脏的细胞凋亡指数明显增加,出现了明显的慢性心力衰竭。SCAD mRNA和蛋白水平在老年自发性高血压大鼠慢性心力衰竭中的表达明显下调,心肌组织ATP含量下降,线粒体膜电位升高,线粒体肿胀程度加大。结论 SCAD在老龄自发性高血压大鼠慢性心力衰竭中明显下调,可能与自发性高血压老龄化导致的心力衰竭密切相关。
Objective To observe the changes of short-chain acyl-CoA dehydrogenase(SCAD)in chronic heart failure(CHF)of elderly spontaneously hypertensive rats,and explore the potential role of SCAD in CHF of elderly spontaneously hypertensive rats.Methods Twenty-month-old male SHR and age-matched WKY control rats were divided into WKY control group and SHR experimental group.Echocardiogram,blood pressure,left ventricular mass index,myocardial fibrosis,apoptosis,the expression of SCAD mRNA and protein,ATP content in myocardial tissue,mitochondrial membrane potential and degree of swelling were measured.Results Compared with WKY control group,cardiac ejection fraction and fractional shortening were reduced,cardiac output was decreased significantly,and the apoptosis index of the heart was increased in SHR group marked with chronic heart failure.The expression of SCAD mRNA and protein was down-regulated in elderly spontaneously hypertensive rats with CHF.The content of ATP in myocardial tissue was decreased,the mitochondrial membrane potential was increased,and the degree of mitochondrial swelling was increased.Conclusion SCAD is obviously down-regulated in chronic heart failure of aged spontaneously hypertensive rats,which may be closely related to heart failure caused by aging of spontaneously hypertensive rats.
